Faeth Therapeutics announced plans to host a virtual key opinion leader (KOL) event on October 6, 2026, focused on multi-node inhibition of the PI3K/AKT/mTOR (PAM) pathway and its potential application in endometrial cancer. The event will feature Marcus Goncalves, MD, PhD, of NYU Langone Health and a scientific co-founder of Faeth, alongside company management. The discussion is expected to provide an overview of the PAM pathway, Faeth’s lead investigational program PIKTOR, and the company’s upcoming Phase 2 readout in endometrial cancer. Management will also discuss translational work supporting the current PIKTOR dose and schedule, prior Phase 1b clinical data and the therapeutic rationale for targeting multiple points within a frequently altered oncogenic pathway. Topline Phase 2 data are anticipated by year-end 2026, making the KOL event an important corporate and clinical-development communication ahead of the planned readout.
PIKTOR Uses Vertical Blockade of the PAM Pathway
PIKTOR is designed to address limitations associated with single-node pathway inhibition by simultaneously targeting PI3K and mTOR. The investigational, all-oral combination pairs serabelisib, a selective PI3K-alpha inhibitor, with sapanisertib, an mTORC1/mTORC2 inhibitor. Faeth is pursuing a vertical pathway-blockade strategy intended to inhibit multiple nodes within the PAM signaling network, which regulates cell growth, survival and metabolism and is frequently dysregulated across human cancers. During the October event, management plans to compare single-node inhibition with PIKTOR’s multi-node approach and review translational findings that informed the clinical development program. The company said published research indicates that PAM pathway alterations are present in approximately 80% of endometrial tumors, supporting the rationale for continued investigation of pathway-directed therapies in this setting.
Endometrial Cancer Program Approaches Phase 2 Readout
Faeth is advancing PIKTOR in a Phase 2 study for patients with second-line advanced endometrial cancer, with topline results expected by the end of 2026. The development program is focused on a setting where treatment options can remain limited following checkpoint inhibitor therapy, creating an opportunity for alternative targeted approaches. The company plans to use the KOL event to discuss the unmet need in endometrial cancer and the clinical rationale behind PIKTOR’s multi-node inhibition strategy. The event will also include discussion of clinical data from an earlier Phase 1b study, providing additional context for the ongoing Phase 2 program. Faeth Expands PIKTOR Development Into Breast Cancer

PIKTOR is being evaluated in the Phase 1b/2 FTH-PIK-101 study, with the first patient dosed in April 2026 and interim data anticipated in 2027. The broader program reflects Faeth’s strategy of applying multi-node inhibition to cancers in which oncogenic signaling pathways contribute to disease progression and treatment resistance.
