Medicus Pharma Ltd. announced a strategic transformation into a precision oncology-led biotechnology company, with CD228V, a second-generation CD228-targeted antibody-drug conjugate (ADC), now representing the company’s principal strategic priority. Medicus plans to concentrate initial CD228V development on melanoma, particularly patients with relapsed or refractory disease, while using an integrated clinical, translational and biomarker strategy to establish clinical proof-of-concept. The program is supported by an active U.S. Investigational New Drug (IND) application and a first-in-human Phase 1 clinical study (NCT06799533). Medicus believes successful proof-of-concept in melanoma could provide a foundation for subsequent development in other CD228-expressing solid tumors, strengthening the company’s repositioning around precision-guided cancer therapeutics.
CD228V Becomes Principal Clinical Development Focus
CD228V is a second-generation ADC targeting melanotransferrin, or CD228, a cell-surface protein expressed across multiple solid tumors. Medicus is initially focusing the program on melanoma, where the company sees an opportunity to evaluate the therapy in a carefully selected patient population with significant unmet medical need. The investigational ADC utilizes Pfizer’s established vedotin linker-payload platform and is supported by prior scientific, translational and clinical development. Beyond melanoma, CD228 expression has been identified across tumor types including squamous non-small cell lung cancer, head and neck squamous cell carcinoma and esophageal cancer, potentially providing opportunities for future indication expansion. Medicus’ strategy is centered on combining clinical development with biomarker and translational analysis to better understand CD228 biology, patient selection and potential therapeutic response as the program progresses through early-stage clinical development.
SkinJect and Teverelix Development Strategies Sharpen
While CD228V becomes the company’s primary focus, Medicus Pharma is refining the development strategies for SkinJect and Teverelix through targeted programs and capital-efficient partnerships. For SkinJect, the company believes the strongest risk-adjusted opportunity is in Gorlin syndrome, rather than the broader sporadic and nodular basal cell carcinoma market. The FDA has authorized initiation of SKNJCT-005, an NDA-enabling registrational Phase 2b study in patients with Gorlin syndrome who develop multiple BCC lesions. Medicus believes the rare-disease focus could provide potential strategic benefits, including opportunities for Orphan Drug Designation and Rare Pediatric Disease designation, as well as rare-disease-oriented pricing. For Teverelix, the company intends to prioritize an optimized approximately 126-patient Phase 2 acute urinary retention program and the PRECISION-E2 Phase 2a study in women with symptomatic endometriosis, while exploring potential partnerships to advance its positioning in advanced prostate cancer.
