BOSTON, Massachusetts — September 8, 2026
Verastem Oncology announced plans to host an investor event and webcast on October 14, 2026, featuring initial efficacy and updated safety and tolerability data for VS-7375, its investigational oral KRAS G12D inhibitor, across pancreatic, lung and colorectal cancers. The presentation will provide a broader clinical assessment of VS-7375 from the ongoing TARGET-D development program, building on clinical data reported in June. Verastem management will be joined by Manuel Hidalgo Medina, M.D., Ph.D., Director of the Gastrointestinal Cancer Center at NYU Langone Health and an investigator in the ongoing VS-7375 clinical trials. The upcoming event represents an important development and investor communication milestone as Verastem advances the program across multiple KRAS G12D-mutated solid tumors and seeks to establish the potential best-in-class profile of its dual ON/OFF KRAS G12D inhibitor.
VS-7375 Targets Both Active and Inactive KRAS G12D
VS-7375 is designed as a potent and selective oral KRAS G12D dual ON/OFF inhibitor, targeting both the active and inactive states of KRAS G12D. The approach is intended to provide more comprehensive pathway inhibition than compounds designed to bind only one state of the protein. KRAS G12D mutations are important drivers across several solid tumors, creating significant demand for targeted therapies capable of suppressing KRAS-driven tumor growth. Verastem is evaluating VS-7375 through the TARGET-D clinical development program, which includes the Phase 1/2 TARGET-D 101 study for dose escalation, expansion and combination development. The company is also advancing three registration-directed Phase 2 studies focused on metastatic pancreatic ductal adenocarcinoma, advanced non-small cell lung cancer and metastatic colorectal cancer.
TARGET-D Program Expands Across Three Major Tumor Types
Verastem is positioning VS-7375 for development across multiple KRAS G12D-mutated cancers, with three Phase 2 trials currently enrolling patients. TARGET-D 201 is evaluating the therapy in metastatic pancreatic ductal adenocarcinoma (PDAC), including the first-line setting, while TARGET-D 202 is studying advanced non-small cell lung cancer (NSCLC), including patients with asymptomatic brain metastases. TARGET-D 203 is evaluating VS-7375 in metastatic colorectal cancer (CRC), also including first-line treatment. This multi-indication development strategy provides Verastem with opportunities to evaluate the molecule across different KRAS G12D-driven tumor settings while building a broader clinical dataset. The company has also received FDA Fast Track Designation for VS-7375 in first-line and previously treated KRAS G12D-mutated PDAC and in previously treated KRAS G12D-mutated unresectable locally advanced or metastatic NSCLC.
Verastem Builds Global Development and Commercialization Strategy
The continued expansion of the TARGET-D program strengthens Verastem Oncology’s broader RAS/MAPK-focused development strategy, with VS-7375 representing a key addition to its pipeline of targeted small-molecule cancer therapies. The molecule originated through Verastem’s collaboration with GenFleet Therapeutics, which is developing VS-7375 as GFH375, while Verastem holds development and commercialization rights outside mainland China, Hong Kong, Macau and Taiwan. The October investor event is expected to provide additional visibility into VS-7375’s clinical activity and tolerability across three major solid tumor types and could help define the next stages of the program. With multiple Phase 2 studies underway, FDA Fast Track Designations secured and broader clinical data scheduled for presentation, Verastem is advancing VS-7375 toward potential registration while expanding its position in KRAS G12D-targeted oncology.
