Pharvaris announced publication of a Pharvaris-funded analysis of the multinational Adelphi Real World HAE Disease Specific Programme in Advances in Therapy, evaluating real-world patterns in the management of hereditary angioedema (HAE). The analysis examined attack frequency and severity, use of long-term prophylaxis (LTP) and on-demand treatment (ODT), treatment adherence, treatment decisions and patient and physician satisfaction. The findings identified continued gaps between guideline-recommended care and real-world treatment experiences despite the availability of multiple prophylactic and on-demand options for HAE.
Treatment Burden Influences Patient Satisfaction and Adherence
The analysis found that treatment satisfaction and decisions to change therapy were influenced by portability, route of administration, perceived efficacy and the likelihood of adverse events. Treatment burden, particularly convenience and how therapies fit into daily life, also affected patient behavior and satisfaction. For on-demand treatment, a substantial proportion of patients did not treat every HAE attack, most commonly because attacks were perceived as insufficiently severe to justify treatments associated with greater burden. Other reasons included lack of treatment availability and pain associated with injectable therapies. For long-term prophylaxis, patients showed a preference for daily oral treatment over biweekly or monthly injections, highlighting the importance of treatment usability and convenience in real-world disease management.
Real-World Study Combines Patient and Physician Perspectives
The analysis used data from the Adelphi Real World HAE Disease Specific Programme collected between January 2023 and January 2024 across the United States, Europe and Japan. The study included 225 physicians, who completed record-based assessments for 1,131 patients with HAE, alongside 279 patients who provided direct self-reported information through questionnaires. This dual approach allowed researchers to evaluate clinical management patterns alongside patient experiences, including treatment behaviors, adherence, satisfaction and quality-of-life impact. Pharvaris noted that the HAE treatment landscape has evolved since the survey period, but said the findings remain relevant to understanding real-world treatment experiences.
Pharvaris Focuses on Oral HAE Treatment Options
The findings reinforce the importance of treatment convenience and patient preferences in managing bradykinin-mediated angioedema. Pharvaris is developing novel oral bradykinin B2 receptor antagonists for conditions including HAE and acquired angioedema due to C1 inhibitor deficiency. The company aims to develop therapies combining injectable-like efficacy with oral administration and a well-tolerated profile for both prevention and treatment of attacks. By highlighting persistent treatment burdens and gaps between recommended care and real-world use, the published analysis provides additional context for the potential role of convenient oral therapies in HAE management.
