NeuroTherapia, Inc. has announced encouraging findings from its Phase 2a clinical trial evaluating NTRX-07, an investigational oral therapy for Alzheimer’s disease (AD), with results presented at the Alzheimer’s Association International Conference (AAIC) 2026 in London. The randomized, double-blind, placebo-controlled study evaluated the safety, tolerability, pharmacokinetics, and exploratory biomarkers of NTRX-07 in 48 patients with mild cognitive impairment (MCI) or mild-to-moderate Alzheimer’s disease. The clinical data demonstrated target engagement, favorable safety, and encouraging biomarker changes consistent with the drug’s proposed anti-neuroinflammatory mechanism of action. The findings provide important evidence supporting further clinical development of NTRX-07 as a potential disease-modifying therapy targeting microglial activation and neuroinflammation, two biological processes increasingly recognized as key contributors to Alzheimer’s disease progression. With positive pharmacodynamic signals observed across imaging and cerebrospinal fluid biomarkers, NeuroTherapia plans to advance the therapy into larger controlled clinical studies.
Phase 2a Trial Demonstrates Target Engagement and Safety
The 28-day multicenter Phase 2a trial successfully confirmed that NTRX-07 was safe and well tolerated, with pharmacokinetic results consistent with previously established therapeutic exposure levels. In addition to evaluating safety, researchers investigated several exploratory imaging and biomarker endpoints designed to assess whether the therapy engages its intended biological targets. Advanced diffusion MRI analyses suggested reductions in neuroinflammation across several brain regions, with some measurements reaching statistical significance while others demonstrated favorable directional trends. Investigators also identified distinct biomarker patterns among treatment responders, particularly within the superior parietal region, supporting the potential biological activity of the investigational therapy in patients with early Alzheimer’s disease.
Biomarker Findings Support Anti-Neuroinflammatory Mechanism
The encouraging neuroimaging results were reinforced by multiple cerebrospinal fluid (CSF) biomarker findings consistent with the proposed mechanism of NTRX-07. Patients receiving treatment demonstrated stable or reduced Neurofilament Light Chain (NfL) levels compared with increases observed in the placebo group, suggesting reduced neuronal injury. Researchers also reported improvements in the Aβ42/Aβ40 ratio, indicating favorable changes in amyloid biology. Additional reductions were observed in inflammatory biomarkers including IL-6 and YKL-40, supporting evidence that NTRX-07 may successfully reduce neuroinflammation by modulating microglial activity. Preclinical studies have previously demonstrated that selective activation of the CB2 receptor restores normal microglial function, promotes amyloid clearance, reduces inflammation, and improves memory and neuronal function, findings now supported by encouraging early clinical observations.
Advancing Disease-Modifying Therapy for Alzheimer’s Disease
The latest Phase 2a data strengthen NeuroTherapia’s strategy of developing oral small-molecule therapies targeting neuroinflammation, an area receiving growing attention in Alzheimer’s research. Unlike therapies that focus primarily on amyloid removal, NTRX-07 seeks to modify disease progression by regulating the brain’s immune response through selective CB2 receptor activation, potentially offering broader neuroprotective benefits. Company leaders emphasized that the combination of a favorable safety profile, pharmacodynamic target engagement, and encouraging biomarker signals provides a strong foundation for larger controlled clinical trials designed to evaluate clinical efficacy. Beyond Alzheimer’s disease, NeuroTherapia also sees potential applications for NTRX-07 across additional central nervous system (CNS) disorders, including Parkinson’s disease, amyotrophic lateral sclerosis (ALS), multiple sclerosis (MS), and other neuroinflammatory conditions. As research continues to highlight the central role of neuroinflammation in neurodegenerative diseases, the Phase 2a findings position NTRX-07 as a promising investigational candidate in the evolving landscape of next-generation Alzheimer’s therapeutics.
