Idorsia Ltd. announced that new analyses from the Phase 3 PRECISION study will highlight the antihypertensive efficacy of aprocitentan in patients with obesity-associated resistant hypertension and across the cardiovascular-kidney-metabolic (CKM) continuum. The findings will be presented in two oral sessions at the American Heart Association Hypertension Scientific Sessions 2026, taking place October 7-11 in Arlington, Virginia. The analyses further support targeting the endothelin pathway in patients with difficult-to-control hypertension, including those with obesity, diabetes and chronic kidney disease.
Aprocitentan Evaluated in Obesity-Associated Resistant Hypertension
A new post hoc analysis of PRECISION evaluated aprocitentan in patients with resistant hypertension who were obese or severely obese, many of whom also had diabetes, chronic kidney disease or albuminuria. The analysis examines the role of the endothelin pathway in obesity-associated hypertension, where increased endothelin-1 activity may contribute to blood pressure elevation and cardiovascular and kidney complications. The findings are intended to provide additional evidence supporting endothelin receptor blockade as a therapeutic approach for resistant hypertension in patients affected by obesity. The results will be presented by Dr. Michael A. Weber during an oral session on October 10.
PRECISION Analysis Explores the CKM Disease Continuum
A second analysis evaluated aprocitentan across different stages of cardiovascular-kidney-metabolic syndrome, examining patients across early and more advanced CKM disease. CKM syndrome reflects the interconnected effects of obesity, diabetes, chronic kidney disease and cardiovascular disease, with hypertension acting as an important driver of disease progression. The analysis provides additional information on the effects of endothelin pathway blockade across the CKM continuum and its potential relevance to blood pressure control and cardiovascular, kidney and metabolic health. The findings will be presented by Dr. John M. Flack in an oral session at AHA Hypertension 2026.
Aprocitentan Advances Treatment of Resistant Hypertension
Aprocitentan is a once-daily oral dual endothelin receptor antagonist that blocks the binding of endothelin-1 to ETA and ETB receptors. The therapy is approved in the United States under the brand name TRYVIO® for systemic hypertension in combination with other antihypertensive drugs in adults who are not adequately controlled on other medicines. It is also approved as JERAYGO™ in the European Union, UK, Switzerland and Canada for resistant hypertension. The pivotal PRECISION Phase 3 study enrolled patients whose systolic blood pressure remained elevated despite treatment with three antihypertensive medicines, including a diuretic. With new data focusing on obesity and CKM disease, Idorsia is expanding the clinical evidence supporting aprocitentan and the role of endothelin pathway inhibition in difficult-to-control hypertension.
