IDEAYA Biosciences announced the initiation of Part 2 monotherapy expansion in its ongoing Phase 1/2 clinical trial evaluating IDE892, a potential best-in-class MTA-cooperative PRMT5 inhibitor, for patients with MTAP-deleted pancreatic ductal adenocarcinoma (PDAC) and non-small cell lung cancer (NSCLC). The milestone follows successful completion of multiple dose-escalation cohorts, where the investigational therapy achieved projected efficacious target exposures with 24-hour EC90 coverage while the maximum tolerated dose (MTD) has not yet been reached. The advancement highlights IDEAYA’s continued progress in developing precision oncology therapies targeting genetically defined cancers with significant unmet medical needs.
IDE892 Designed for Precision Targeting and Combination Therapy
IDE892 has been engineered with approximately 1,400-fold selective MTA-PRMT5 cooperative binding compared with SAM-PRMT5 binding, a feature intended to maximize tumor selectivity while minimizing off-target effects. The investigational therapy also demonstrated favorable drug-like properties, including no time-dependent inhibition across the seven major cytochrome P450 enzymes and a CYP3A4 IC50 greater than 45 micromolar, making it an attractive candidate for future combination regimens. The company is simultaneously advancing combination dose-escalation studies of IDE892 with IDE397, its MAT2A inhibitor, in MTAP-deleted NSCLC and PDAC, while preparing to initiate a first-patient-in study combining IDE892 with Roche’s pan-RAS inhibitor during the second half of 2026. These programs are designed to exploit synthetic lethal vulnerabilities created by MTAP deletion, a genomic alteration found in up to 40% of pancreatic cancers and approximately 15% of NSCLC cases, where no approved targeted therapies currently exist.
Broad MTAP Pipeline Targets Pancreatic Cancer Challenges
According to IDEAYA, the company is building one of the industry’s most comprehensive MTAP-deletion precision oncology pipelines. Beyond IDE892 and the Phase 2-stage IDE397, the company is advancing a potential first-in-class CDKN2A-targeted program through preclinical toxicology studies, with an Investigational New Drug (IND) application planned for the first half of 2027. Since MTAP deletion frequently co-occurs with CDKN2A loss and KRAS mutations, particularly in pancreatic cancer, IDEAYA plans to develop rational combination strategies addressing tumor heterogeneity and resistance mechanisms. The company also announced plans to host a dedicated MTAP/CDKN2A, KRAS and Pancreatic Cancer R&D Day in the fourth quarter of 2026 to discuss its expanding research strategy and future clinical development plans.
Clinical Expansion Strengthens IDEAYA’s Precision Oncology Strategy
The initiation of the Part 2 monotherapy expansion represents a significant development for IDEAYA as it advances IDE892 toward broader clinical evaluation in patients with MTAP-deleted solid tumors. By combining highly selective PRMT5 inhibition, favorable pharmacologic characteristics, and multiple combination strategies targeting complementary cancer pathways, the company aims to address one of oncology’s largest genetically defined unmet needs. With ongoing Phase 1/2 studies, planned combination trials, and additional pipeline programs targeting CDKN2A and KRAS-driven cancers, IDEAYA continues to strengthen its position in the rapidly evolving field of precision medicine, with several important clinical milestones expected through 2026 and 2027.
