San Francisco, California, USA, November 3, 2026
Thryv Therapeutics Inc., a clinical-stage biotechnology company focused on developing innovative treatments for cardiovascular and inherited cardiac disorders, has announced $30 million in Series B financing to advance its lead drug candidate, THRV-1268, into a new clinical study targeting heart failure. The financing was led by InClin, Inc., a global contract research organization, with continued participation from existing investors. The announcement marks an important milestone in Thryv’s cardiovascular drug development programme, particularly its efforts to address significant unmet medical needs in patients with heart failure and inherited cardiac rhythm disorders. The proceeds will support the launch of ASPIRE-HF, a Phase 2a clinical study evaluating THRV-1268 in adults with heart failure with reduced ejection fraction (HFrEF) and prolonged corrected QT interval (QTc). The company expects the study to open for patient enrollment in early 2027. The financing announcement also follows the completion of enrollment in the first efficacy cohort of the company’s Wave II Phase 2/3 clinical study investigating THRV-1268 in patients with Long QT Syndrome. These developments highlight the company’s strategy of advancing a targeted therapeutic approach that addresses both electrical instability and structural changes associated with cardiovascular disease.
Thryv Secures $30 Million Series B Financing
The $30 million Series B financing represents a significant funding milestone for Thryv Therapeutics as it expands its clinical development activities in cardiovascular medicine. The financing was led by InClin, Inc., which brings both investment support and clinical research operational capabilities to the collaboration. InClin has operations in San Mateo, California, and Sydney, Australia, and has been working with Thryv to establish clinical operations in Australia to support the upcoming ASPIRE-HF programme. The investment will primarily fund the Phase 2a study through a prespecified analysis of safety and efficacy, including changes in QTc measurements and established biomarkers associated with heart failure risk. Existing investors supporting the financing include Fonds de solidarité FTQ, Amplitude Ventures, Investissement Québec, Lumira Ventures, adMare BioInnovations, CTI Life Sciences Fund and Alexandria Venture Investments. The participation of these investors reflects continued financial support for Thryv’s development of selective serum and glucocorticoid-inducible kinase 1 (SGK1) inhibitors. The company is focusing on SGK1 as a potential therapeutic target because of its involvement in biological processes associated with cardiac electrical activity, inflammation, fibrosis and structural remodeling. The new funding is expected to provide resources to advance clinical research and evaluate the therapeutic potential of THRV-1268 in additional cardiovascular indications.
ASPIRE-HF Phase 2a Study Targets Heart Failure
The major clinical development supported by the financing is ASPIRE-HF (Addressing SGK1-driven Prolonged QT and Impaired Remodeling in Heart Failure), a randomized, double-blind, placebo-controlled Phase 2a study. The trial will evaluate THRV-1268 in adults with symptomatic heart failure with reduced ejection fraction and prolonged QTc who are receiving guideline-directed medical therapy. Participants will be assigned to one of two dose levels of THRV-1268 or placebo over a 24-week treatment period. A prespecified early efficacy assessment is planned at Week 6. The study will investigate the safety, pharmacokinetics and potential effects of the investigational treatment on markers of arrhythmic risk, cardiac function and cardiac remodeling. HFrEF is a serious cardiovascular condition in which the heart cannot pump sufficient blood to meet the body’s requirements. Patients with prolonged QTc may also face increased risks associated with abnormal electrical activity and potentially dangerous cardiac arrhythmias. Thryv is investigating whether selective SGK1 inhibition could address several biological processes contributing to these complications. According to the company, human genetic evidence, preclinical findings and earlier Phase 1 observations support further investigation of this therapeutic approach. However, the clinical benefits of THRV-1268 in heart failure have not yet been established.
Wave II Long QT Syndrome Study Completes First Cohort
Alongside the planned heart failure programme, Thryv reported that its Wave II Phase 2/3 clinical study of THRV-1268 in Long QT Syndrome has completed enrollment of its first efficacy cohort. The company expects initial efficacy data in early 2027. During the third quarter of 2026, the study’s Independent Data Monitoring Committee (IDMC) reviewed available safety and pharmacokinetic information and recommended that the trial continue without modification. Thryv also reported that no serious adverse events had been observed at the time of the announcement. The company has submitted a protocol amendment to its U.S. FDA Investigational New Drug application to include patients with Long QT Syndrome Type 3 (LQT3), expanding the programme beyond its earlier focus. The study is now open to participants aged 15 years and older. THRV-1268 is an investigational, highly selective SGK1 inhibitor being developed as a potential treatment for inherited cardiac channelopathies and cardiometabolic disorders. Further clinical evidence will be necessary to establish its safety and effectiveness across these conditions. Thryv also plans to present new research on SGK1 inhibition in cardiometabolic heart failure at the American Heart Association Scientific Sessions 2026, scheduled for November 6–9 in Chicago, Illinois. Overall, the $30 million financing, planned ASPIRE-HF study and progress in Wave II represent important steps in advancing the company’s cardiovascular drug development pipeline.
