OSAKA, Japan, and CAMBRIDGE, Massachusetts, October 15, 2026
Takeda (TSE: 4502; NYSE: TAK) announced the simultaneous availability of ORZEYFUL (oveporexton) in Japan and China for the treatment of narcolepsy type 1 (NT1), giving physicians in both countries access to a first-in-class orexin receptor 2 (OX2R) agonist. Discovered by Takeda scientists in Japan, the oral tablet is designed to restore orexin signaling and address the underlying orexin deficiency associated with NT1. The launches follow regulatory approvals within approximately one month of one another by China’s National Medical Products Administration, the U.S. Food and Drug Administration and Japan’s Ministry of Health, Labour and Welfare. Takeda said ORZEYFUL is the lead medicine in its broader orexin development franchise, which includes investigational programs for additional sleep-wake disorders and other conditions involving orexin biology. The company expects to launch ORZEYFUL in the United States in November 2026, following completion of Drug Enforcement Administration scheduling, while regulatory submissions are underway in other markets worldwide.
First-in-Class Orexin Agonist Targets the Biology of NT1
Narcolepsy type 1 is a chronic, rare neurological disease associated with orexin deficiency, which disrupts the regulation of wakefulness and sleep. Symptoms can include excessive daytime sleepiness, cataplexy, disrupted nighttime sleep, sleep paralysis, hallucinations and cognitive difficulties. These symptoms may affect patients’ independence, education, employment, social interactions and overall quality of life. According to Takeda’s announcement, people with narcolepsy also experience approximately twice the healthcare resource utilization of control populations, including more emergency department visits, hospitalizations, outpatient appointments and physician consultations. ORZEYFUL selectively stimulates the orexin receptor 2, with the aim of restoring signaling that is deficient in people with NT1. By targeting this biological pathway, the treatment is designed to address a broader range of disease manifestations rather than focusing exclusively on individual symptoms. Takeda describes the medicine as the first therapy designed to restore orexin signaling and target the underlying biological cause of NT1. The availability of the oral treatment in Japan and China marks a new commercial stage for the company’s orexin research program and expands access to the medicine in two major markets..
Phase 3 Results Show Improvements Across Multiple Symptoms
Takeda reported that landmark Phase 3 clinical studies demonstrated improvements in wakefulness, sleepiness, cataplexy, disease severity and quality-of-life measures among people with NT1 receiving ORZEYFUL compared with placebo. According to the announcement, the studies showed consistent, clinically meaningful and statistically significant improvements across doses over a 12-week period, with reported p-values below 0.001 for the measures described. Improvements were observed at the earliest assessed time point for each measure and were sustained throughout the study period. The company also reported that ORZEYFUL was generally well tolerated, with a safety profile consistent across clinical studies conducted to date. The most common side effects identified in the announcement include insomnia, urinary urgency, urinary frequency and excessive saliva. These findings underpin the company’s positioning of oveporexton as a treatment intended to address multiple aspects of NT1 through selective activation of OX2R. Takeda International President Giles Platford said the company sees potential for the treatment to change how NT1 is managed in Japan and China, noting that patients have reported improvements in daily functioning and quality of life. The reported results reflect the clinical studies described by the company; individual treatment outcomes and tolerability may vary.
Global Expansion Supports Takeda’s Broader Orexin Franchise
The launches in Japan and China are part of Takeda’s strategy to develop medicines targeting orexin biology across multiple conditions. The company said ORZEYFUL is approved in China for adolescents aged 16 and older and adults with NT1, and in Japan and the United States for adults with NT1. The planned U.S. launch is expected next month, subject to completion of DEA scheduling. Takeda is also pursuing regulatory submissions in other markets. Beyond oveporexton, the company’s orexin portfolio includes TAK-360, an investigational oral orexin agonist being studied for NT1, narcolepsy type 2 and idiopathic hypersomnia, as well as TAK-495. Takeda is exploring the potential relevance of orexin agonists to additional sleep-wake disorders and other indications involving respiration, mood and metabolism. The company is headquartered in Japan and operates across approximately 80 countries and regions, with research and development activities spanning gastrointestinal and inflammatory conditions, rare diseases, plasma-derived therapies, oncology, neuroscience and vaccines. ORZEYFUL’s initial launches represent a commercial milestone for Takeda’s neuroscience portfolio, while the broader franchise reflects its continuing efforts to translate orexin research into potential treatment options for conditions involving disrupted sleep-wake regulation.
