Polyrizon Ltd. announced enrollment of the first patient in its U.S. clinical study of NASARIX (PL-14), an investigational drug-free nasal hydrogel being developed for seasonal allergic rhinitis (SAR). The milestone marks Polyrizon’s transition into clinical-stage development and represents the first evaluation of NASARIX in patients. The multicenter study is designed to assess the efficacy, safety and tolerability of the investigational Allergy Blocker under seasonal allergen exposure.
NASARIX Study Evaluates Allergy Symptoms
The prospective, randomized, single-blind study is expected to enroll approximately 120 adults across five U.S. clinical sites, with participants randomized 1:1 to receive NASARIX Allergy Blocker or a saline nasal spray comparator. The primary efficacy endpoint will evaluate the difference in mean daily reflective Total Nasal Symptom Score (rTNSS) between treatment groups during the first 14 days. Safety assessments will include treatment-emergent adverse events and local and overall tolerability.
Drug-Free Hydrogel Forms Nasal Barrier
NASARIX is a non-pharmacological, mucoadhesive hydrogel nasal spray designed to form a physical barrier inside the nasal cavity. The barrier is intended to reduce contact between inhaled airborne allergens and nasal mucosal tissue, potentially helping reduce symptoms associated with seasonal allergic rhinitis. The product is based on Polyrizon’s proprietary Capture and Contain™ (C&C) hydrogel technology, which uses naturally occurring building blocks to create a thin shield-like containment barrier following nasal administration.
Polyrizon Advances Capture and Contain Platform
The NASARIX clinical study represents the first clinical-stage application of Polyrizon’s Capture and Contain technology, following product development, preclinical evaluation, manufacturing preparation and regulatory activities. Beyond NASARIX, the company is exploring C&C technology for nasal delivery applications, including potential barrier-based approaches against viruses and allergens. Polyrizon is also developing its earlier-stage Trap and Target™ (T&T) technology for intranasal delivery of active pharmaceutical ingredients, expanding the company’s broader nasal drug-delivery platform.
