Ionis Pharmaceuticals, Inc. announced that new clinical data supporting its severe hypertriglyceridemia (sHTG) treatment programs will be presented at the European Society of Cardiology (ESC) Congress 2026, taking place August 28–31 in Munich, Germany. The presentations will feature one-year data for TRYNGOLZA® (olezarsen) from its long-term extension study, alongside Phase 1 results for ION775, an investigational next-generation siRNA targeting APOC3. The upcoming scientific presentations are designed to highlight Ionis’ continued development of RNA-targeted approaches for patients with very high triglyceride levels and reinforce the company’s expanding cardiovascular pipeline.
Ionis Highlights TRYNGOLZA Long-Term sHTG Data
A key presentation at ESC 2026 will provide one-year efficacy and safety data for olezarsen in patients with severe hypertriglyceridemia. TRYNGOLZA was approved by the U.S. Food and Drug Administration in June 2026 as the first and only approved treatment for adults with sHTG to reduce triglycerides and the risk of acute pancreatitis. The treatment is an RNA-targeted therapy designed to reduce production of apoC-III, a liver-produced protein involved in triglyceride metabolism. The long-term data will add to the clinical evidence supporting olezarsen in patients with sHTG, a condition characterized by triglyceride levels of 500 mg/dL or higher and associated with an increased risk of potentially serious acute pancreatitis. An indication extension application for TRYNGOLZA in sHTG is also under review by the European Medicines Agency, with Sobi holding commercialization rights in the European Union.
Ionis Presents ION775 Next-Generation siRNA Data
Ionis will also present 12-month Phase 1 data for ION775, an investigational long-acting siRNA designed to target APOC3 and potentially provide less frequent dosing than TRYNGOLZA. The presentation will focus on single-dose results in participants with moderate hypertriglyceridemia and will assess the durability of the investigational therapy over 12 months. ION775 is currently being evaluated in a Phase 2 study for severe hypertriglyceridemia, making the ESC presentation an important clinical update for Ionis’ next-generation cardiometabolic program. The company is investigating whether the long-acting siRNA approach could ultimately support semiannual or less frequent administration, potentially offering a different treatment profile for patients requiring long-term triglyceride management..
Ionis Expands Cardiovascular RNA Medicine Portfolio
In addition to its sHTG programs, Ionis and AstraZeneca will present data from the Phase 3 CARDIO-TTRansform trial of eplontersen in patients with transthyretin-mediated amyloid cardiomyopathy (ATTR-CM). Planned presentations include detailed Phase 3 analyses, a subgroup analysis involving patients receiving stabilizer therapy and an independent academic meta-analysis evaluating silencer therapies used with stabilizers. Together, the ESC 2026 presentations demonstrate the breadth of Ionis’ cardiovascular RNA medicine strategy, spanning triglyceride metabolism and transthyretin-mediated disease. For sHTG specifically, the combination of commercially approved TRYNGOLZA, ongoing long-term data and the development of ION775 illustrates Ionis’ effort to establish a sustained pipeline of APOC3-targeted therapies aimed at addressing persistent cardiovascular and pancreatitis risks associated with severe hypertriglyceridemia.
