SOUTH SAN FRANCISCO, Calif. — October 18, 2026
Genentech, a member of the Roche Group, announced plans to present new oncology data spanning lung, breast and other cancers at the 2026 European Society for Medical Oncology (ESMO) Annual Meeting, taking place October 23–27 in Madrid, Spain. The presentations include two late-breaking abstracts and findings from multiple late-stage clinical programs, highlighting advances in targeted therapies, endocrine treatment and antibody-drug conjugates (ADCs). Key updates will feature divarasib in KRAS G12C-mutated non-small cell lung cancer (NSCLC), giredestrant in estrogen receptor (ER)-positive, HER2-negative breast cancer, and tambotatug pelitecan (Tam-Peli) in advanced solid tumors. According to Genentech, the data reflect its focus on improving treatment outcomes while considering tolerability and patients’ quality of life. Levi Garraway, M.D., Ph.D., chief medical officer and head of Global Product Development at Genentech, said the company’s presentations reflect its efforts to advance treatments for cancers with significant unmet medical needs.
Divarasib Advances in KRAS G12C-Mutated Lung Cancer
A key presentation will feature results from the Phase III Krascendo 1 trial, evaluating divarasib against the first-generation KRAS G12C inhibitors sotorasib and adagrasib in previously treated patients with locally advanced or metastatic KRAS G12C-mutated NSCLC. Genentech reported that the study demonstrated superior efficacy for divarasib compared with these current treatment options, supporting its potential as a next-generation targeted therapy. The company described Krascendo 1 as the only global head-to-head trial evaluating a next-generation KRAS G12C inhibitor against approved first-generation inhibitors. The findings are scheduled for presentation as late-breaking abstract LBA5 during Presidential Symposium II on October 25, from 4:30 to 4:42 p.m. Central European Time. Genentech will also present Krascendo 3, a randomized Phase III trial evaluating divarasib against immunotherapy or observation in patients with resected stage II–IIIB KRAS G12C-positive NSCLC. This study-in-progress presentation is scheduled for October 26. Together, the trials reflect the company’s efforts to investigate targeted treatment strategies across both previously treated advanced disease and earlier-stage lung cancer.
Giredestrant Data Focus on Efficacy and Patient Experience
Genentech will present several analyses of giredestrant, an investigational oral, next-generation selective estrogen receptor degrader (SERD) and full antagonist being studied in ER-positive, HER2-negative breast cancer. More than 90% of cases of this breast cancer subtype are diagnosed at an early stage, according to the company. Patient-reported outcomes from the Phase III lidERA Breast Cancer trial suggest that giredestrant’s improved efficacy compared with standard endocrine therapy was achieved without compromising tolerability or health-related quality of life. These findings are scheduled for a rapid oral presentation on October 23. Additional data will include exploratory biomarker analyses from the Phase III evERA Breast Cancer trial, evaluating giredestrant plus everolimus in advanced ER-positive, HER2-negative disease following treatment with a CDK4/6 inhibitor. Updated interim results from the MORPHEUS Breast Cancer study will also examine giredestrant combined with inavolisib in locally advanced or metastatic ER-positive, HER2-negative breast cancer. The presentations will provide further information on the investigational treatment’s potential across different stages of disease and treatment settings.
Tam-Peli and Other Programs Expand the Solid Tumor Pipeline
The ESMO program will also feature tambotatug pelitecan (Tam-Peli; YL201), an investigational next-generation ADC targeting B7-H3. Designed to deliver potent antitumor activity while limiting off-target toxicity, Tam-Peli is being studied across multiple solid tumor types. A late-breaking Phase III interim analysis from the TAISHAN-301 trial will evaluate Tam-Peli against chemotherapy in heavily pretreated patients with recurrent or metastatic nasopharyngeal carcinoma. The presentation is scheduled for October 25. Additional findings will cover interstitial lung disease analyses from Phase I/II studies in advanced solid tumors and Phase I efficacy and safety data in locally advanced or metastatic NSCLC. These results build on findings from the Phase III TAISHAN-302 study in second-line small cell lung cancer, presented at the World Conference on Lung Cancer in September, which met its three specified endpoints of overall survival, progression-free survival and objective response rate at the first interim analysis. Roche is developing and commercializing Tam-Peli outside mainland China, Hong Kong and Macau, with plans to initiate global trials across multiple tumor types. Genentech will also present data involving other oncology programs, including atezolizumab-based treatment studies and the investigational c-Met-targeting ADC YL211. The ESMO presentations collectively highlight the breadth of the company’s oncology research, while the investigational programs remain subject to further clinical evaluation.
