CytomX Therapeutics has announced a significant expansion of its strategic research collaboration and licensing agreement with Regeneron Pharmaceuticals, strengthening their joint effort to develop next-generation conditionally activated bispecific cancer therapies. The expanded partnership builds on a collaboration originally established in 2022 and leverages the combined strengths of CytomX’s PROBODY® therapeutic platform and Regeneron’s Veloci-Bi® bispecific antibody technology. Under the updated agreement, CytomX will receive a $37 million target selection payment for two newly selected development programs, while Regeneron gains the option to nominate up to six additional future targets. Including target nomination payments, development, regulatory, and commercial milestones, the expanded collaboration now carries a potential value of approximately $4 billion, highlighting the growing industry focus on innovative immunotherapy technologies designed to improve the precision and effectiveness of cancer treatment.
Expanding the Potential of Conditional Bispecific Immunotherapy
The collaboration is focused on developing conditionally activated bispecific antibodies that remain inactive in healthy tissues and become activated only within the tumor microenvironment. This strategy combines CytomX’s proprietary masking technology with Regeneron’s expertise in bispecific antibody engineering to create highly targeted immunotherapies designed to improve safety and efficacy. Traditional T-cell engaging therapies can sometimes trigger unwanted immune activity outside tumors, limiting their therapeutic window.
By utilizing the PROBODY platform, researchers aim to minimize off-target effects while maintaining potent anti-tumor activity. The approach has the potential to unlock previously challenging cancer targets and broaden the application of immunotherapy across a wider range of solid tumors. If all available options are exercised and milestone objectives are achieved, CytomX could receive up to approximately $4 billion in aggregate payments, in addition to tiered royalties on future product sales.
