Consano Bio, Inc. announced that the U.S. Food and Drug Administration (FDA) has granted Fast Track Designation to C-1101, the company’s lead investigational platelet-derived biologic being developed for chronic, painful lumbosacral radiculopathy (LSR), commonly known as chronic sciatica. The designation follows the FDA’s recent clearance of Consano Bio’s Investigational New Drug (IND) application for C-1101 and supports continued clinical development of the potentially disease-modifying, non-opioid biologic. The company is currently evaluating C-1101 in a Phase 1 clinical study as it seeks to develop a treatment that could address the underlying biology of nerve injury rather than solely managing pain symptoms.
FDA Fast Track Highlights Unmet Need in Chronic Sciatica
Chronic lumbosacral radiculopathy is a debilitating condition caused by irritation or damage to spinal nerve roots in the lower back, resulting in pain that can radiate into the leg along with numbness and weakness. Consano Bio highlights the substantial clinical and socioeconomic burden associated with chronic LSR and notes that there are currently no FDA-approved pharmaceutical therapies specifically indicated for chronic sciatica. The condition can persist for years and, in some patients, may progress to lasting nerve damage and disability. Chronic painful LSR is also associated with significant healthcare utilization and can contribute to opioid initiation and prolonged opioid use. The FDA’s Fast Track Designation is intended to facilitate the development and expedited review of therapies designed to treat serious conditions where significant unmet medical needs remain. Consano Bio said the designation was supported by the unmet need in LSR and a comprehensive nonclinical data package submitted regarding C-1101’s clinical potential.
C-1101 Advances in Phase 1 Clinical Study
Consano Bio is advancing C-1101 through a multicenter, randomized, controlled, double-blind Phase 1 clinical trial evaluating the investigational therapy in symptomatic patients with chronic painful LSR. The C-1101-101 study is being conducted through three sequential dose-escalation cohorts, with participants receiving a single epidural injection of the investigational biologic. The study initially began enrolling patients in Australia and is expanding into the United States following FDA clearance of the company’s IND application in July 2026. The ongoing Phase 1 program is primarily intended to evaluate the safety, tolerability and clinical characteristics of C-1101 as dose escalation progresses. The transition into U.S. clinical development represents an important milestone for Consano Bio as it builds the clinical evidence required to determine whether the investigational biologic can advance into later-stage development for chronic sciatica.
Novel Biologic Designed to Support Nerve Repair
C-1101 is described by Consano Bio as a novel platelet-derived multi-protein biologic therapeutic designed to modulate inflammation and support cellular repair at the site of nerve injury. The investigational product contains consistent concentrations of cytokines, growth factors and matrix proteins, which are intended to help activate the body’s natural healing response. Delivered through an epidural injection, C-1101 is designed to provide concentrated levels of these proteins directly at the site associated with nerve injury. This mechanism distinguishes the candidate from conventional approaches that primarily focus on reducing pain symptoms. If successful in clinical development, C-1101 could represent a new therapeutic strategy for patients with chronic LSR by potentially addressing biological processes contributing to persistent nerve dysfunction. However, the therapy remains investigational, and its safety and efficacy have not yet been established by the FDA. The Fast Track designation and ongoing Phase 1 study mark important steps in the company’s efforts to advance a potential non-opioid pharmaceutical treatment for chronic sciatica and address a substantial unmet need in orthopedic and neurological care.
