Boehringer Ingelheim has initiated a Phase II clinical trial evaluating BI 3034701, an investigational GLP-1/GIP/NPY2 triple receptor agonist, in people living with obesity and overweight. The candidate is designed to simultaneously target multiple biological pathways involved in appetite regulation, metabolism, and body weight control, strengthening the company’s expanding cardiometabolic and obesity pipeline.
Triple Receptor Agonist Designed to Address Obesity Through Multiple Mechanisms
BI 3034701 is a potential first-in-class therapy that activates the GLP-1, GIP, and NPY2 receptors. While GLP-1 and GIP receptor activation supports satiety, weight reduction, and metabolic regulation, NPY2 receptor agonism is being investigated for its ability to influence central hunger signaling and appetite control. By combining these complementary mechanisms, the investigational therapy aims to provide a differentiated treatment approach for the heterogeneous nature of obesity and its associated metabolic complications.
Phase II Trial Builds on Favorable Phase I Safety Findings
The newly launched Phase II study will evaluate the efficacy, safety, and optimal dosing of BI 3034701 in adults with obesity and overweight. Advancement into Phase II follows Phase I studies demonstrating a generally favorable safety and tolerability profile. The trial represents an important milestone in Boehringer Ingelheim’s strategy to develop innovative therapies that address obesity as a key driver of cardiovascular, renal, and metabolic diseases.
Expanding Cardiometabolic Pipeline with Next-Generation Therapies
BI 3034701 was originally developed in collaboration with Gubra, with Boehringer Ingelheim retaining responsibility for global clinical development and commercialization. The program complements the company’s broader obesity portfolio, which includes survodutide, a dual glucagon/GLP-1 receptor agonist currently in Phase III development, along with additional next-generation injectable and oral therapies targeting obesity and related metabolic disorders. Through its diversified pipeline, Boehringer Ingelheim aims to deliver more personalized treatment options that address both the biological and behavioral drivers of obesity while improving long-term cardiometabolic outcomes.
