AIM ImmunoTech Inc. highlighted recent biotech media coverage surrounding its development strategy for Ampligen® (rintatolimod) in metastatic pancreatic cancer as the company advances the Phase 2 DURIPANC clinical trial toward topline results expected in the first quarter of 2027. The company said recent coverage has focused on the scientific rationale for using Ampligen to address immune resistance in pancreatic cancer and its continued evaluation in combination with the checkpoint inhibitor Imfinzi® (durvalumab). Pancreatic cancer remains difficult to treat in part because of its immunosuppressive tumor microenvironment, which can limit the effectiveness of immune-based approaches. AIM is evaluating whether Ampligen’s immune-modulating activity can help alter this environment and support coordinated immune responses in patients with metastatic disease.
Ampligen Targets Immune Resistance in Pancreatic Cancer
Ampligen is being developed as an immune-modulating therapy designed to stimulate immune activity through selective activation of Toll-like receptor 3 (TLR3). AIM describes Ampligen as a double-stranded RNA (dsRNA) and highly selective TLR3 agonist with broad-spectrum activity observed in clinical studies. The company’s pancreatic cancer strategy is based on the potential for immune modulation to address mechanisms that contribute to resistance to cancer immunotherapy. Pancreatic tumors can develop an environment that suppresses immune activity, creating challenges for treatments that depend on effective immune-cell engagement. AIM is therefore evaluating Ampligen in combination with durvalumab, an immune checkpoint inhibitor marketed as Imfinzi, in the Phase 2 DURIPANC program. According to the company, interim observations from the study have included changes in the immune system suggesting coordinated activation of both innate and adaptive immune responses. AIM has also reported that the combination has been well tolerated and that patients have consistently reported a high quality of life during treatment. These observations remain part of an ongoing clinical program and are being evaluated alongside the study’s planned clinical outcomes.
DURIPANC Advances Toward Topline Results
The Phase 2 DURIPANC trial has completed final Ampligen dosing, representing a key clinical development milestone for AIM ImmunoTech. The company expects topline results from the study in the first quarter of 2027. DURIPANC is evaluating Ampligen in combination with durvalumab in patients with metastatic pancreatic cancer, with exploratory biomarker analyses intended to provide additional information about potential treatment response and immune changes. AIM said these analyses could help the company understand which patients may benefit from the treatment strategy and inform the design of a planned pivotal clinical trial. The upcoming results will therefore provide clinical information beyond the previously reported immune-system observations, including data that could help determine the next stage of development for the Ampligen program. The company has positioned the DURIPANC readout as an important opportunity to evaluate its approach to overcoming immune resistance in a disease where treatment options remain limited.
Earlier Ampligen Experience Supports Development Strategy
AIM is also drawing on experience from a Netherlands-approved Named Patient Program (NPP) involving Ampligen as maintenance treatment in pancreatic cancer. The program included 57 patients with radiologically stable pancreatic cancer after FOLFIRINOX, according to the company. AIM reported encouraging findings involving overall survival, safety and quality of life from this experience. While the NPP was not a randomized Phase 2 clinical trial and therefore cannot establish comparative efficacy, the company is using the experience as part of the broader rationale for continued clinical development. The DURIPANC program is intended to provide a more structured clinical evaluation of Ampligen in combination with durvalumab and to generate additional information on treatment response and biomarkers. With final Ampligen dosing completed and topline data expected in Q1 2027, AIM ImmunoTech is moving toward an important clinical readout that could help define the future development path for Ampligen in metastatic pancreatic cancer.
