Whitehawk Therapeutics presented new real-world analyses supporting PTK7 as a durable antibody-drug conjugate (ADC) target in patients with EGFR wild-type non-small cell lung cancer (NSCLC) at the IASLC 2026 World Conference on Lung Cancer (WCLC) in Seoul, South Korea. The analyses evaluated PTK7 expression and target stability in tumor samples collected from patients after treatment with standard-of-care therapies. The findings provide additional biological support for continued development of PTK7-targeted ADCs, including Whitehawk’s investigational HWK-007, and suggest that PTK7 expression may remain sufficiently stable following conventional treatment to support continued therapeutic targeting. The company said the findings are particularly relevant to the development of ADCs for patients with EGFR wild-type NSCLC, where additional targeted treatment approaches are needed.
Real-World Data Support PTK7 Target Stability
Whitehawk’s analyses examined PTK7 expression in real-world tumor samples from patients with EGFR wild-type NSCLC who had received standard-of-care treatment. The company reported that PTK7 expression remained broadly consistent after exposure to commonly used therapies, supporting the hypothesis that the target can remain available for therapeutic intervention as the disease progresses. The analysis is important for ADC drug development because successful targeted delivery depends on the continued presence of the selected antigen on tumor cells. If expression of a target decreases substantially following standard therapy, subsequent ADC treatment could potentially become less effective. The real-world findings presented by Whitehawk instead provide evidence that PTK7 expression can persist after treatment, supporting its potential utility as a target in later-line NSCLC. The company also reported evidence of PTK7 target engagement in clinical samples from patients treated with HWK-007. Target engagement refers to the ability of a therapeutic agent to interact with its intended biological target, providing an important pharmacodynamic measure during early clinical development. These findings complement the expression analyses by providing evidence that the PTK7 target can be accessed by the investigational ADC in patients.
HWK-007 Advances PTK7-Directed ADC Research
HWK-007 is an investigational ADC being developed by Whitehawk Therapeutics to target PTK7, a cell-surface protein expressed across multiple solid tumors, including subsets of lung cancer. The therapeutic strategy combines a targeting antibody with a cytotoxic payload, with the objective of selectively delivering the payload to PTK7-expressing tumor cells. The company’s clinical program is currently evaluating HWK-007 in a Phase 1 clinical trial designed to characterize the candidate’s safety, tolerability, pharmacokinetics and preliminary antitumor activity. Early-stage studies such as this are essential for determining appropriate dosing and establishing whether the biological mechanism translates into measurable clinical responses. Whitehawk’s WCLC analyses do not establish the clinical efficacy of HWK-007. Instead, they provide translational and biomarker evidence that supports the underlying rationale for targeting PTK7. Further clinical data will be required to determine whether PTK7-directed ADC treatment can produce meaningful and durable tumor responses in patients. The focus on EGFR wild-type NSCLC is also clinically relevant. While molecularly selected EGFR-mutated tumors can be treated with targeted EGFR therapies, patients without actionable EGFR alterations have fewer precision-treatment options. Identifying additional tumor-associated targets such as PTK7 could therefore expand the range of targeted therapies available for this population.
Findings Strengthen ADC Development Strategy
The presentation at WCLC 2026 adds to the growing body of research investigating ADCs against novel targets in solid tumors. ADC technology has gained significant momentum in oncology because it can combine the selectivity of antibody-based targeting with the potent cytotoxic activity of a small-molecule payload. For PTK7, demonstrating durable target expression and target engagement is an important component of the development strategy. The Whitehawk analyses suggest that PTK7 remains detectable in tumor tissue following standard treatment, potentially allowing an ADC directed against the target to remain relevant in later stages of disease. However, real-world biomarker analyses have inherent limitations compared with prospective randomized clinical trials. The findings primarily establish a biological rationale for continued investigation, rather than demonstrating improved progression-free or overall survival. Additional prospective clinical data will be needed to establish the safety and efficacy profile of HWK-007 and determine which patients are most likely to benefit. Whitehawk’s latest WCLC presentation therefore represents a meaningful drug-development milestone for its PTK7 program.
