Waltham, Mass., September 2, 2026
TScan Therapeutics, Inc. announced a strategic reorganization to prioritize its in vivo-engineered TCR-T cell therapy program for solid tumors, advancing two therapeutic candidates targeting PRAME and MAGE-A4 into IND-enabling studies. The company plans to share preclinical data in Q1 2027, file its first Investigational New Drug (IND) application in Q3 2027, and potentially begin Phase 1 clinical development in Q4 2027. TScan believes its in vivo approach could address key limitations associated with traditional ex vivo autologous TCR-T therapies, including patient-specific manufacturing complexity, treatment delays and the need for lymphodepletion.
TScan Advances PRAME and MAGE-A4 TCR-T Candidates
The company’s updated strategy centers on developing in vivo-engineered TCR-T therapies for solid tumors, initially as single-target therapies with the longer-term goal of developing multiplexed approaches. TScan has now advanced its first two candidates, directed against PRAME and MAGE-A4, into IND-enabling studies. The company believes this approach could simplify the delivery of engineered T-cell therapies by addressing manufacturing and logistical challenges associated with conventional autologous cell therapy. TScan expects to provide additional preclinical data in Q1 2027, followed by its planned first IND filing later in 2027.
TScan Reports Updated TSC-101 Data but Pauses Phase 3 Enrollment
Alongside the strategic shift, TScan reported updated data from Cohort C of the Phase 1 ALLOHA study evaluating TSC-101 in hematologic malignancies. All 13 patients currently being tracked demonstrated complete donor chimerism, including two patients who had previously experienced relapse and subsequently converted to complete donor chimerism after additional treatment. However, TScan has paused further enrollment in the Phase 3 ALLOHA-2 study because of insufficient capital to complete the trial. The company will continue following the seven patients already enrolled in the treatment arm and the 13 Cohort C patients while seeking strategic partners for its hematology program.
TScan Restructures Operations and Extends Runway Into Q4 2027
The reorganization includes a workforce reduction of approximately 75%, elimination of the company’s internal manufacturing organization and a significant reduction in its research footprint. TScan expects the restructuring to generate approximately $55 million in cumulative cost savings through the end of 2027. The company said its cash, cash equivalents and marketable securities as of June 30, 2026, are expected to fund planned operations into Q4 2027. TScan also intends to pursue partnerships for its hematologic malignancy and autoimmune programs, while concentrating its available resources on the development of in vivo TCR-T cell therapy for solid tumors.
