Tiziana Life Sciences announced new quantitative PET imaging results from the third patient with Multiple System Atrophy (MSA) to complete dosing in its ongoing Phase 2 clinical trial evaluating intranasal foralumab, a fully human anti-CD3 monoclonal antibody. The imaging analysis demonstrated reductions in inflammatory activity within disease-relevant brain regions, including up to a 34% reduction in standardized uptake value (SUV) and a 26% reduction in standardized uptake value ratio (SUVR) in the basal ganglia and cerebellar white matter. The findings are consistent with results previously reported in the first two treated patients, providing additional evidence supporting the therapy’s potential to reduce neuroinflammation in this rare neurodegenerative disease.
Funding to Accelerate Clinical Development and Pipeline Expansion
Quantitative PET scan comparisons performed before and after treatment demonstrated meaningful reductions in radiotracer uptake across brain regions commonly affected by MSA. The third patient exhibited up to 34% reduction in SUV and 26% reduction in SUVR, closely matching the reductions observed in the first two patients, who recorded approximately 35% SUV and 24% SUVR improvements. According to investigators, these reproducible imaging findings provide encouraging evidence of biological activity and suggest that intranasal foralumab may reduce neuroinflammatory processes associated with disease progression. Researchers noted that additional quantitative analyses and correlation with clinical outcomes will be important to further validate these observations.
Foralumab Designed to Modulate Immune Response Through Nasal Delivery
Intranasal foralumab is a fully human anti-CD3 monoclonal antibody designed to induce immune modulation through mucosal tolerance mechanisms rather than systemic immune suppression. The investigational therapy aims to reduce inflammation while avoiding the systemic toxicities commonly associated with traditional anti-CD3 therapies. Beyond MSA, foralumab has also demonstrated encouraging findings in patients with non-active secondary progressive multiple sclerosis (na-SPMS) and moderate Alzheimer’s disease, supporting broader development across neuroinflammatory and neurodegenerative disorders.
Program Continues as Company Evaluates Additional Patients
Multiple System Atrophy is a rare, progressive neurodegenerative disorder characterized by autonomic dysfunction, parkinsonism, and cerebellar ataxia, with no approved disease-modifying therapies currently available. Tiziana plans to continue enrolling and evaluating additional patients in the Phase 2 study to further characterize intranasal foralumab’s ability to reduce neuroinflammation and determine whether the observed imaging improvements translate into meaningful clinical benefit. The company believes the consistent PET imaging findings across multiple patients strengthen the rationale for continued development of the therapy in MSA and other neurological diseases.
