Satellite Bio announced that the U.S. Food and Drug Administration (FDA) has granted Rare Pediatric Disease (RPD) designation to its investigational off-the-shelf liver therapy SB-101 for the treatment of urea cycle disorders (UCDs). The designation recognizes the urgent need for innovative therapies targeting these rare, life-threatening inherited metabolic disorders, which primarily affect infants and children and currently have no curative treatment available during the critical first weeks of life. SB-101 is being developed as a first-in-class liver cell therapy designed to restore essential metabolic liver function by delivering healthy liver cells capable of replacing lost hepatic activity. The FDA designation supports the program’s clinical development while highlighting the significant unmet medical need in patients with severe early-onset UCDs. Satellite Bio plans to initiate a Phase 1/2 clinical trial of SB-101 during 2026, marking an important milestone in advancing regenerative therapies for pediatric liver diseases.
SB-101 Designed to Restore Liver Function in Infants with Severe UCDs
Urea cycle disorders are devastating inherited metabolic diseases caused by defects in the body’s ability to remove ammonia, leading to toxic accumulation that can rapidly result in encephalopathy, seizures, coma, organ failure, severe neurological injury, and death. Current treatment options remain limited and largely supportive, with mortality exceeding 25% and approximately half of surviving patients experiencing long-term cognitive impairment. SB-101 aims to address the underlying cause of these disorders by delivering healthy functional liver cells that restore essential metabolic processes rather than simply managing symptoms. Manufactured using Satellite Bio’s proprietary scalable cell expansion platform, the therapy is designed as an off-the-shelf, cryopreserved product that is readily available when urgently needed, offering potential advantages in availability, manufacturing consistency, and cost compared with conventional individualized cell therapies.
