Resolution Therapeutics announced the expansion of its pipeline for inflammatory and fibrotic diseases through the development of an in vivo Regenerative Macrophage Therapy (RMT) platform. The company has signed Material Transfer Agreements (MTAs) with multiple leading lipid nanoparticle (LNP) technology providers to evaluate advanced delivery systems for its proprietary mRNA payloads. The initiative is designed to complement the company’s lead autologous macrophage therapy RTX001, which is currently being evaluated in the Phase 1/2 EMERALD clinical trial for patients with end-stage liver disease and decompensated liver cirrhosis. Together, the two development programs are intended to broaden patient access to regenerative macrophage therapies while accelerating innovation in chronic inflammatory and fibrotic diseases.
LNP-Based In Vivo Platform Aims to Accelerate Therapy Development
Resolution Therapeutics will systematically assess multiple best-in-class lipid nanoparticle formulations using a fast-fail development strategy, allowing the company to evaluate several delivery technologies simultaneously. This parallel approach is designed to rapidly identify the most effective LNP platform capable of delivering proprietary mRNA payloads directly to the myeloid compartment without triggering an inflammatory macrophage response. Alongside LNP optimization, the company is refining its proprietary payloads to maximize therapeutic efficacy. Successful pairing of an optimized LNP delivery system with an effective payload could pave the way for lead candidate nomination studies and significantly shorten development timelines for in vivo regenerative macrophage therapies.
In Vivo Program Complements RTX001 Clinical Development
The newly announced in vivo platform builds upon the company’s lead asset, RTX001, an autologous engineered macrophage therapy currently being investigated in the ongoing EMERALD Phase 1/2 clinical trial. The study is evaluating RTX001 in patients with end-stage liver disease who have experienced decompensated liver cirrhosis, a condition where liver transplantation often remains the only available treatment option. Interim clinical data from EMERALD are expected later this year. By advancing both autologous cell therapy and scalable in vivo therapeutic platforms simultaneously, Resolution aims to develop complementary treatment options capable of addressing a wider range of patients affected by chronic inflammatory and fibrotic disorders.
Regenerative Macrophage Therapy Targets Tissue Repair Across Multiple Diseases
Regenerative Macrophage Therapy (RMT) is designed to restore the body’s natural healing processes by reprogramming macrophages to reduce both inflammation and fibrosis, two major drivers of chronic disease progression. Unlike therapies targeting a single biological pathway, RMT seeks to regenerate damaged tissues by modifying the disease microenvironment through macrophages’ dual anti-inflammatory and anti-fibrotic functions.
