Rani Therapeutics Holdings, Inc., a clinical-stage biotherapeutics company pioneering oral biologic drug delivery, has announced a research and development collaboration with PegBio Co., Ltd. to expand its obesity and metabolic disease pipeline by leveraging the company’s proprietary RaniPill® oral delivery platform. The strategic collaboration will focus on developing oral formulations for multiple novel obesity and metabolic disease candidates from PegBio’s proprietary portfolio, with the goal of advancing the most promising molecules into future clinical development. The agreement significantly strengthens Rani Therapeutics’ obesity franchise, broadening its pipeline beyond existing programs while reinforcing the company’s mission to transform injectable biologic therapies into convenient oral medicines. As obesity continues to emerge as one of the fastest-growing therapeutic markets worldwide, the partnership positions both companies to accelerate innovation across multiple differentiated mechanisms of action while expanding treatment options for patients living with chronic metabolic disorders.
Strategic Collaboration Broadens Next-Generation Obesity Pipeline
Under the terms of the collaboration, Rani Therapeutics and PegBio will jointly conduct preclinical research and development using the RaniPill® capsule platform to evaluate multiple proprietary obesity and metabolic disease molecules selected from PegBio’s growing pipeline. The objective is to identify the most promising candidates capable of progressing into clinical development while demonstrating the feasibility of delivering complex biologics through an oral dosage form rather than traditional injections.
The collaboration substantially expands Rani’s growing obesity portfolio, complementing ongoing internal development programs while providing access to multiple novel therapeutic candidates targeting diverse biological pathways involved in obesity and metabolic disease. Company leadership noted that expanding collaborations across multiple candidates reflects increasing confidence in the versatility of the RaniPill® platform and its potential to unlock new opportunities for oral biologic therapies addressing significant unmet medical needs within obesity management.
RaniPill® Platform Aims to Transform Injectable Biologics into Oral Medicines
The proprietary RaniPill® capsule represents one of the most innovative oral drug delivery technologies currently under clinical development, designed to replace subcutaneous injections and intravenous infusions with convenient oral administration of biologic medicines. The technology has successfully demonstrated encouraging safety, tolerability, bioavailability, and pharmacokinetic performance across multiple preclinical and clinical studies, validating its ability to deliver large biologic molecules that traditionally require injection. The collaboration builds upon Rani’s expanding obesity franchise, including RT-114, a GLP-1/GLP-2 dual agonist currently undergoing Phase I clinical evaluation through an existing partnership with ProGen.
In addition, the company has completed several promising preclinical studies involving semaglutide and other obesity candidates, generating pharmacokinetic, pharmacodynamic, bioavailability, and weight-loss data comparable to injectable formulations. These findings further support the potential of the RaniPill® technology to significantly improve patient convenience, treatment adherence, and long-term outcomes by offering effective oral alternatives to injectable biologic therapies.
Advancing Innovation in Obesity and Metabolic Disease Therapeutics
The collaboration reflects the rapidly evolving landscape of obesity drug development, where demand for innovative therapies continues to accelerate alongside increasing global prevalence of obesity, diabetes, and metabolic disorders. By combining PegBio’s proprietary therapeutic pipeline with Rani Therapeutics’ advanced oral biologics platform, the companies aim to create differentiated treatment options capable of addressing multiple biological mechanisms associated with obesity while eliminating the burden of routine injections.
Beyond expanding its own development pipeline, the agreement further strengthens Rani Therapeutics’ position as a leader in oral biologic drug delivery, supporting broader applications across immunology, endocrinology, gastroenterology, and other chronic diseases where injectable biologics remain the standard of care. As development progresses, the collaboration has the potential to accelerate the next generation of oral obesity medicines, supporting improved patient access, greater treatment convenience, and continued innovation in metabolic disease therapeutics while advancing the future of precision biologic drug delivery through the RaniPill® platform.
