NEW YORK, Aug. 4, 2026
Pathos AI announced a collaboration and co-exclusive licensing agreement with AstraZeneca to advance AZD4241, an investigational oral estrogen receptor alpha (ERα) PROTAC (proteolysis-targeting chimera), into early clinical development for patients with estrogen receptor-positive, HER2-negative (ER+/HER2-) breast cancer. Under the agreement, Pathos AI will lead the early clinical development of AZD4241, leveraging its proprietary Foundry AI platform to optimize patient selection, trial design, and dose selection. The collaboration strengthens Pathos AI’s expanding oncology pipeline while combining AstraZeneca’s drug discovery expertise with AI-driven clinical development to accelerate the advancement of targeted therapies for breast cancer patients whose disease progresses on standard endocrine treatment.
AZD4241 Targets Estrogen Receptor Degradation with PROTAC Technology
AZD4241 is a preclinical investigational oral PROTAC therapy specifically engineered to degrade the estrogen receptor (ERα), targeting both wild-type and mutated forms of the receptor. Mutations in the estrogen receptor are found in approximately 30% to 40% of patients whose ER+/HER2- breast cancer progresses despite hormone therapy, representing a major challenge in current treatment. Rather than simply blocking receptor activity, PROTAC technology promotes selective degradation of the disease-causing protein, offering a differentiated therapeutic strategy that may overcome endocrine resistance. By advancing AZD4241 into clinical development, the companies aim to evaluate whether this novel mechanism can provide improved treatment options for patients with advanced hormone receptor-positive breast cancer.
Foundry AI Platform Will Optimize Clinical Development
Pathos AI will utilize its proprietary Foundry platform, an AI-native drug development system, to guide every aspect of AZD4241’s early clinical program. The platform deploys thousands of AI agents powered by the company’s Pathos Oncology Foundation Model to analyze large-scale biological, clinical, and real-world datasets. These AI capabilities are designed to identify optimal patient populations, improve dose selection, accelerate trial enrollment, and enhance overall study design while continuously monitoring clinical performance. By integrating artificial intelligence throughout drug development, Pathos AI aims to reduce development timelines and improve the likelihood of identifying patients most likely to benefit from targeted therapies, addressing one of oncology’s most significant clinical development challenges.
Collaboration Expands Pathos AI’s Precision Oncology Pipeline
The collaboration with AstraZeneca expands Pathos AI’s oncology pipeline to three major therapeutic assets spanning four different tumor types. AZD4241 joins pocenbrodib, currently being developed for metastatic castration-resistant prostate cancer (mCRPC) and relapsed/refractory multiple myeloma (RRMM), along with DO-2, which targets MET-altered non-small cell lung cancer (NSCLC). Every development program within the company’s portfolio has been identified, prioritized, or accelerated using the Foundry AI platform, reinforcing Pathos AI’s strategy of integrating artificial intelligence across the entire drug development process. The new agreement highlights the growing role of AI-powered precision oncology in advancing innovative cancer therapies from discovery through clinical development.
