NewAmsterdam Pharma Company N.V. announced that it will present multiple clinical analyses highlighting the potential of obicetrapib as a novel oral, low-dose therapy for the prevention of early Alzheimer’s disease at the Alzheimer’s Association International Conference (AAIC) 2026, taking place July 12–15, 2026, in London, United Kingdom. The presentations will feature data from the company’s Phase 3 BROADWAY trial, exploring the relationship between CETP inhibition, Alzheimer’s disease biomarkers, and cardiovascular risk. The analyses focus on the effects of obicetrapib on plasma p-tau217, one of the leading biomarkers associated with Alzheimer’s pathology, while also examining additional biomarkers and genetic factors that may help identify patients most likely to benefit from future preventive treatment strategies. The findings further expand the potential clinical applications of obicetrapib beyond its primary cardiovascular development program.
BROADWAY Trial Analyses Highlight Alzheimer’s Biomarkers
The AAIC 2026 presentations include one oral presentation and three scientific posters examining data from the BROADWAY Phase 3 trial. Researchers analyzed 1,535 patients, including 367 ApoE4 carriers, to evaluate changes in plasma biomarkers associated with Alzheimer’s disease after 12 months of treatment with obicetrapib. The company reported statistically significant reductions in p-tau217, a key biomarker linked to Alzheimer’s pathology, compared with placebo in both the overall study population and the genetically defined ApoE4 carrier subgroup. Additional analyses demonstrated favorable trends in other Alzheimer’s-related biomarkers, including neurofilament light chain (NFL), glial fibrillary acidic protein (GFAP), p-tau181, and the Aβ42/40 ratio, supporting further investigation of CETP inhibition as a potential disease prevention strategy.
Obicetrapib Continues to Advance in Late-Stage Clinical Development
Obicetrapib is a once-daily, oral CETP inhibitor being developed primarily for patients with elevated low-density lipoprotein cholesterol (LDL-C) who remain inadequately controlled with existing lipid-lowering therapies or cannot tolerate standard treatment. Across multiple Phase 2 studies and Phase 3 clinical trials, including BROADWAY, BROOKLYN, TANDEM, and PREVAIL, the investigational therapy has consistently demonstrated statistically significant LDL-C reductions while maintaining a safety profile comparable to placebo. The PREVAIL Phase 3 cardiovascular outcomes trial, which enrolled more than 9,500 patients, continues to evaluate whether obicetrapib can reduce major adverse cardiovascular events (MACE). The emerging Alzheimer’s biomarker analyses represent an important extension of the drug’s scientific potential into neurodegenerative disease prevention.
Clinical Research Expands Beyond Cardiovascular Disease
The upcoming AAIC 2026 presentations demonstrate NewAmsterdam Pharma’s broader clinical research strategy of exploring the biological connections between cardiovascular disease and Alzheimer’s disease. By investigating the impact of CETP inhibition on biomarkers associated with neurodegeneration, the company aims to determine whether improving lipid metabolism may also influence the progression of Alzheimer’s pathology, particularly in genetically susceptible individuals such as ApoE4 carriers. While the Alzheimer’s analyses were conducted as a predefined subset of the BROADWAY trial and were not originally designed as a dedicated Alzheimer’s efficacy study, the encouraging biomarker findings provide scientific support for additional clinical investigation. As obicetrapib advances through late-stage cardiovascular development, these exploratory analyses may help shape future clinical trials evaluating its potential role in preventing or delaying the onset of Alzheimer’s disease.
