Mighty Therapeutics, a commercial-stage biotechnology company focused on therapies targeting mitochondrial dysfunction, and a research team from the University of Washington (UW) led by David Marcinek, Ph.D., have been named a Milestone 2 Awardee in the $101 million XPRIZE Healthspan competition. The joint team was selected based on pilot-study data evaluating elamipretide, Mighty’s mitochondria-targeted peptide, in older adults. As one of 10 Milestone 2 Awardees, the team will receive a $1 million award and advance to the final phase of the competition, where teams must conduct rigorous controlled clinical trials designed to evaluate interventions intended to improve healthy aging.
Elamipretide Advances Mitochondrial Aging Research
The pilot study provides data that will help guide the design of a larger, controlled clinical trial evaluating whether elamipretide can improve measures of healthy aging. The planned study is expected to use a randomized, double-blind, placebo-controlled design and will investigate whether targeting mitochondrial function can improve muscle, cognitive, and immune function in older adults. The research builds on the hypothesis that mitochondrial dysfunction contributes to age-related decline and multiple disease processes. According to the research team, improving mitochondrial energetics could potentially address biological mechanisms associated with reduced physical function and quality of life as people age. The recognition from XPRIZE Healthspan provides additional support for advancing this research into a more rigorous clinical evaluation.
Mitochondria-Targeted Therapy Moves Toward Clinical Testing
Elamipretide is designed to bind to cardiolipin, a lipid that plays an important role in maintaining mitochondrial structure and function. By interacting with cardiolipin, the therapy is intended to improve mitochondrial integrity and support the cell’s ability to produce energy. Mighty Therapeutics is developing elamipretide for several rare and age-related conditions, including dry age-related macular degeneration (dry AMD), a major cause of vision loss among older adults. The company is also continuing development of the therapy in mitochondrial diseases. The research program reflects a broader effort to investigate whether therapies that directly address mitochondrial dysfunction can have applications across diseases associated with impaired cellular energy production.
XPRIZE Healthspan Competition Enters Final Phase
The Mighty Therapeutics and University of Washington team is now entering the final phase of the XPRIZE Healthspan competition, which requires finalist teams to evaluate their interventions through rigorous placebo-controlled clinical trials. The competition’s final clinical testing phase is scheduled to continue through 2029, with the overall competition expected to culminate in 2030. The judging panel may award up to $81 million to the team or teams whose interventions demonstrate the ability to improve muscle, cognitive, and immune function in older adults. The $1 million Milestone 2 award therefore represents both recognition of the preliminary research and an opportunity to advance the program into a larger clinical investigation.
