Fosun Pharma and AriBio announced the signing of an exclusive global option agreement for the development, manufacturing, registration, and commercialization of AR1001, an investigational oral therapy for Alzheimer’s disease (AD) currently being evaluated in a global Phase 3 clinical trial. The agreement significantly expands the companies’ existing collaboration and strengthens Fosun Pharma’s international presence in neurodegenerative disease therapeutics and innovative CNS drug development.
Under the terms of the agreement, Fosun Pharma will pay AriBio a US$60 million option fee and retain the right to exercise a global licensing option during the agreed exercise period. If exercised, the transaction could include up to US$180 million in upfront and regulatory milestone payments, with additional commercial milestone payments tied to annual net sales thresholds exceeding US$2.5 billion.
The collaboration extends beyond the companies’ previous regional partnership, which had granted Fosun Pharma commercialization rights for AR1001 in mainland China, Hong Kong, Macao, and selected ASEAN markets. The new agreement broadens the partnership into major international markets including the United States, Europe, and Japan, positioning AR1001 as a potential global Alzheimer’s disease therapy candidate.
AR1001 Advances as Potential Disease-Modifying Alzheimer’s Therapy
AR1001 is a once-daily oral phosphodiesterase-5 (PDE-5) inhibitor designed as a potential disease-modifying treatment for Alzheimer’s disease. The therapy is currently being studied in the global POLARIS-AD Phase 3 clinical trial, which has enrolled more than 1,500 patients across the United States, Europe, China, South Korea, and additional international regions. Enrollment was completed in 2025, and top-line trial results are expected in 2026.
According to AriBio, AR1001 has demonstrated therapeutic activity across multiple biological pathways associated with Alzheimer’s disease progression. The investigational therapy has shown potential neuroprotective effects, inhibition of tau phosphorylation, suppression of inflammatory responses, and improved cerebral blood flow in preclinical and clinical evaluations to date.
The companies stated that existing data suggest favorable blood-brain barrier penetration and an encouraging safety profile in patients ranging from mild cognitive impairment to mild Alzheimer’s dementia. Researchers believe the therapy could offer a differentiated oral treatment option in a disease area where effective disease-modifying therapies remain limited despite growing global demand.
