ROCKVILLE, Md., October 15, 2026
MacroGenics, Inc. (NASDAQ: MGNX), a clinical-stage biopharmaceutical company developing antibody-based cancer therapies, announced that the first patient has been dosed in the Phase 1 clinical trial of MGC030, an investigational antibody-drug conjugate (ADC) designed to target an undisclosed antigen expressed across several solid tumor types. The first-patient dosing milestone advances MacroGenics’ early-stage oncology pipeline and marks its third ADC in active clinical development. The open-label, multicenter study is designed to assess MGC030’s safety, tolerability, pharmacokinetics, immunogenicity and preliminary antitumor activity in patients with advanced solid tumors. The trial will also explore biomarkers that may be associated with these outcomes. Eligible participants include patients with relapsed or refractory, unresectable, locally advanced or metastatic cancers, including urothelial, colorectal, gastric or gastroesophageal junction, esophageal and pancreatic cancers. The dose-escalation portion is expected to enroll up to 54 participants, with the possibility of further enrollment at selected dose levels. According to the company, MGC030 has demonstrated antitumor activity in multiple preclinical models and an encouraging safety and tolerability profile in cynomolgus monkey toxicology studies. However, its safety and efficacy in humans remain under investigation.
Phase 1 Study Evaluates Safety and Preliminary Activity
The MGC030 Phase 1 trial uses a combination of dose escalation, dose optimization and cohort expansion to characterize the investigational therapy in patients with advanced solid tumors. The study’s primary objectives include evaluating safety and tolerability while examining pharmacokinetics, which describe how the drug moves through the body, and immunogenicity, which assesses potential immune responses against the therapeutic molecule. Researchers will also evaluate preliminary antitumor activity and explore biomarkers that may help identify associations with treatment outcomes. The study is open-label and multicenter, and it includes patients whose cancers have relapsed or are refractory to prior treatment or whose tumors are unresectable, locally advanced or metastatic. The planned tumor types include urothelial, colorectal, gastric and gastroesophageal junction, esophageal and pancreatic cancers. The dose-escalation phase will enroll up to 54 participants, with the potential to expand enrollment at selected dose levels. MacroGenics has not disclosed initial clinical results or a projected timeline for data reporting in the announcement. The study is intended to generate early clinical evidence to guide further evaluation of MGC030, including assessment of tolerability, drug exposure and potential antitumor effects across the selected patient populations.
ADC Design Combines Targeted Delivery with Cytotoxic Payload
MGC030 is an investigational ADC directed against an undisclosed antigen expressed across several solid tumors. MacroGenics stated that there are currently no approved therapeutics directed against this target. The candidate incorporates Synaffix’s proprietary ADC technology and the SYNtecan T™ linker-payload, which uses an exatecan-based topoisomerase I inhibitor as its cytotoxic component. The linker is cleavable, and the molecule has a drug-to-antibody ratio (DAR) of 4, meaning that an average of four payload molecules are attached per antibody molecule. ADCs are designed to combine the target-recognition properties of an antibody with the cell-killing activity of a cytotoxic payload. In MGC030’s case, the target antigen is not publicly identified in the announcement, limiting further assessment of its tumor distribution or potential patient-selection strategy. MacroGenics reported that MGC030 showed potent antitumor activity in multiple in vivo preclinical models and an encouraging safety and tolerability profile in cynomolgus monkey toxicology studies. These findings provide preclinical support for clinical evaluation but do not establish that the candidate will demonstrate similar effects or an acceptable safety profile in humans.
MGC030 Expands MacroGenics’ Clinical ADC Pipeline
MacroGenics said the first-patient dosing milestone builds on progress across its antibody-drug conjugate portfolio, with MGC030 becoming the company’s third ADC in active clinical development. Senior Vice President and Chief Scientific Officer Ezio Bonvini, M.D., said preclinical studies had shown encouraging antitumor activity and that the Phase 1 study would deepen the company’s understanding of MGC030’s potential in selected advanced solid tumors. The broader development approach combines MacroGenics’ antibody engineering capabilities with technology platforms used to generate investigational therapies across oncology. The company also has strategic collaborations with pharmaceutical and biotechnology organizations to advance its pipeline. The ongoing MGC030 trial will provide the initial human data needed to characterize the candidate’s safety, tolerability, pharmacokinetics, immunogenicity and preliminary antitumor activity. Further development decisions will depend on the results of the clinical study. The trial is registered on ClinicalTrials.gov under identifier NCT07822399. MGC030 remains investigational and has not been established as a safe or effective treatment for any cancer indication.
