InxMed Co., Ltd., a clinical-stage biotechnology company focused on overcoming cancer treatment resistance, has announced the publication of encouraging clinical results in The Lancet Respiratory Medicine demonstrating the promising anticancer activity of ifebemtinib (IN10018) in combination with garsorasib (D-1553) for the treatment of first-line KRAS G12C-mutant non-small cell lung cancer (NSCLC). The published findings originate from the company’s Phase Ib/II clinical trial, where the all-oral, chemotherapy-free combination regimen delivered impressive efficacy, durable responses, prolonged progression-free survival, and a manageable safety profile. The publication represents a significant scientific milestone for InxMed and further validates its strategy of targeting focal adhesion kinase (FAK) to overcome resistance mechanisms associated with KRAS G12C-targeted therapies, supporting the continued advancement of the combination into a global Phase III confirmatory study.
Clinical Trial Demonstrates Exceptional Response Rates and Survival Benefits
The published study evaluated 33 treatment-naïve patients with KRAS G12C-mutant NSCLC who received ifebemtinib (100 mg once daily) together with garsorasib (600 mg twice daily). After a median follow-up of 21.5 months, investigators observed highly encouraging clinical outcomes. The combination achieved a confirmed objective response rate (cORR) of 82% across all enrolled patients and 87% among evaluable patients, while tumor shrinkage was observed in every evaluable patient. Among responding patients, the median duration of response (DOR) was not reached, highlighting the therapy’s ability to deliver durable clinical benefit.
The study also reported an impressive median progression-free survival (mPFS) of 22.3 months, with 12-month and 18-month progression-free survival rates of 67.9% and 58.2%, respectively. Overall survival data also remained encouraging, with a median overall survival (OS) of 27.8 months, while many patients continued treatment at the time of analysis. Importantly, the combination maintained a manageable safety profile, with most treatment-emergent adverse events classified as Grade 1 or Grade 2, and no treatment-related deaths or treatment discontinuations reported.
FAK Inhibition Offers a Novel Strategy to Overcome KRAS G12C Resistance
The publication also highlights InxMed’s translational research demonstrating that KRAS G12C inhibition alone activates the FAK-YAP signaling pathway, promoting adaptive resistance, tumor survival, and fibrotic changes within the tumor microenvironment. By combining ifebemtinib, a highly selective FAK inhibitor, with garsorasib, researchers successfully interrupted this resistance pathway, potentially extending treatment responses beyond what current KRAS G12C inhibitors can achieve alone.
These findings represent an important advance for patients with KRAS G12C-mutant NSCLC, where no approved first-line KRAS G12C-targeted combination therapy currently exists. Based on these encouraging clinical data, ifebemtinib has already received Breakthrough Therapy Designation (BTD) from China’s National Medical Products Administration (NMPA) for first-line KRAS G12C-mutant NSCLC. InxMed is currently conducting the randomized Phase III confirmatory trial (NCT07174908), which is designed to establish this chemotherapy-free oral regimen as a potential new standard of care for patients with advanced lung cancer.
