NEW YORK – August 26, 2026
Immuneering Corporation (Nasdaq: IMRX) highlighted its continued focus on first-line metastatic pancreatic cancer following the U.S. FDA’s approval of a new treatment for patients in the second-line setting. The company is advancing atebimetinib in combination with chemotherapy through the global randomized Phase 3 MAPKeeper 301 trial, which is currently enrolling patients with previously untreated metastatic pancreatic cancer. Immuneering said the development strategy is focused on improving survival while maintaining patient strength and reducing treatment-related burden.
Immuneering Focuses on First-Line Pancreatic Cancer
MAPKeeper 301 (NCT07562152) is a global, randomized pivotal study evaluating atebimetinib plus chemotherapy in patients with first-line metastatic pancreatic cancer. More than 35 trial locations are already listed for the study, reflecting the company’s ongoing clinical development program. Atebimetinib is an oral, once-daily investigational MEK inhibitor designed under Immuneering’s Deep Cyclic Inhibitor approach. The company is developing the therapy with the goal of controlling tumor growth while potentially supporting patients’ ability to maintain body weight and tolerate treatment. The Phase 3 program represents the next clinical test of this strategy in a larger randomized population.
Phase 2a Data Support Continued Clinical Development
Immuneering previously reported results from an open-label, single-arm Phase 2a study involving 55 patients with first-line metastatic pancreatic cancer who received atebimetinib 320 mg once daily plus modified gemcitabine/nab-paclitaxel (mGnP). At the April 24, 2026 data cutoff, the study reported a median overall survival of 17.3 months, with a median follow-up of 11.6 months. The company also reported that 84% of evaluable patients maintained or gained weight at three months, an important measure in a disease frequently associated with substantial weight and muscle loss. Among treatment-related adverse events of Grade 3 or higher occurring in at least 10% of patients, anemia occurred in 16% and neutropenia in 18%, with both identified as chemotherapy-related.
Atebimetinib Strategy Moves Toward Phase 3 Evaluation
The ongoing MAPKeeper 301 trial now serves as the key clinical development program for atebimetinib in first-line metastatic pancreatic cancer. Immuneering is positioning the investigational therapy around three objectives associated with its Deep Cyclic Inhibitor platform: durable tumor control, preservation of body mass and improved tolerability. The company’s broader pipeline focuses on cancers driven by RAS, RAF and other MAPK signaling pathways, with atebimetinib representing its lead clinical-stage program. While the Phase 2a findings provide encouraging signals for further investigation, the single-arm study cannot establish comparative efficacy; the randomized Phase 3 trial will be critical for determining whether the approach can deliver a meaningful benefit over current treatment strategies in first-line metastatic pancreatic cancer.
