IDEAYA Biosciences and Roche have entered into a strategic clinical collaboration to evaluate a novel targeted treatment combination for patients with MTAP-deleted, RAS-mutant pancreatic ductal adenocarcinoma (PDAC), one of the most aggressive and difficult-to-treat forms of cancer. The collaboration will investigate IDE892, IDEAYA’s investigational Phase 1 MTA-cooperative PRMT5 inhibitor, in combination with RG6505, Roche’s Phase 1 pan-RAS inhibitor, with the goal of improving outcomes for patients who currently have no approved targeted treatment options. The announcement highlights growing momentum in precision oncology, where therapies are increasingly being developed to address specific genetic alterations that drive tumor growth. Given that MTAP deletion occurs in up to 40% of pancreatic ductal adenocarcinoma cases, the collaboration has the potential to address a substantial patient population facing limited therapeutic choices and poor long-term survival outcomes.
Precision Oncology Collaboration Targets High-Unmet-Need Patients
Pancreatic ductal adenocarcinoma remains one of the deadliest cancers worldwide, with most patients diagnosed at advanced stages and facing limited treatment options. Researchers have identified MTAP deletion as a common genetic alteration in PDAC, frequently occurring alongside RAS mutations that promote aggressive tumor behavior. The new collaboration seeks to exploit this biological vulnerability through a dual-targeted strategy. IDE892 was specifically designed as a potential best-in-class PRMT5 inhibitor, targeting synthetic lethality pathways associated with MTAP-deleted tumors.
Roche’s RG6505 is being developed as a pan-RAS inhibitor capable of addressing a broad range of RAS-driven cancers. By combining these two targeted therapies, investigators hope to achieve deeper and more durable anti-tumor responses than either approach could deliver independently. The collaboration reflects a growing trend toward combination precision medicine strategies that attack multiple cancer-driving mechanisms simultaneously to overcome resistance and improve patient outcomes.
Clinical Development Program Expands MTAP-Deletion Strategy
Under the agreement, IDEAYA will sponsor and lead the clinical trial evaluating the combination, while Roche will provide RG6505 for the study. Both companies will participate in joint governance to oversee trial execution and future development decisions. IDE892 has already demonstrated robust tumor regression in MTAP-deleted preclinical models and is currently being evaluated in a Phase 1 dose-escalation study involving patients with MTAP-deleted solid tumors. The newly announced trial will expand this development strategy by specifically targeting MTAP-deleted, RAS-mutant pancreatic cancer patients.
IDEAYA also plans to explore additional combination approaches involving IDE397, its proprietary MAT2A inhibitor, across pancreatic cancer, non-small cell lung cancer (NSCLC), and other solid tumor types. The collaboration framework includes the possibility of evaluating a future triplet regimen consisting of IDE892, RG6505, and IDE397, further strengthening the company’s MTAP-deletion-focused oncology portfolio.
Potential New Treatment Approach for Pancreatic Cancer
The collaboration arrives at a critical time for pancreatic cancer research, as the need for more effective targeted therapies remains urgent. Nearly all MTAP-deleted pancreatic tumors harbor co-occurring RAS mutations, creating a biologically defined patient population that may benefit from precision medicine approaches. Scientists believe that combining a PRMT5 inhibitor with a pan-RAS inhibitor could potentially address multiple tumor survival pathways simultaneously, improving response rates and extending treatment durability.
Importantly, no approved targeted therapies currently exist specifically for patients with MTAP-deleted, RAS-mutant pancreatic cancer. As a result, successful clinical development of this combination could represent a significant advancement in personalized cancer treatment. The partnership also reinforces IDEAYA’s broader mission of developing genetically tailored oncology therapies that leverage synthetic lethality and biomarker-driven patient selection to improve clinical outcomes.
