CAMBRIDGE, UK, July 22, 2026
Healx, an AI-powered clinical-stage biotechnology company, has announced that the first patient has been dosed in its Phase 1/2 clinical trial evaluating HLX-4310, an investigational oral combination therapy designed to prevent metastatic recurrence in relapsed and refractory sarcomas, with a primary focus on osteosarcoma. The milestone marks the first clinical validation of Healx’s broader rare oncology strategy, which combines artificial intelligence-driven drug discovery, rapid translational research, and innovative therapeutic combinations to address cancers with limited treatment options. The study will evaluate HLX-4310, which combines a class I selective histone deacetylase (HDAC) inhibitor with an mTOR inhibitor, in adolescents and adults with advanced sarcoma. The program is being conducted in collaboration with the National Pediatric Cancer Foundation’s Sunshine Projectâ„¢, highlighting a shared commitment to accelerating innovative therapies for patients facing aggressive and difficult-to-treat cancers.
Phase 1/2 Trial Targets Metastatic Recurrence
Unlike many conventional oncology therapies that primarily focus on shrinking established tumors, HLX-4310 has been developed to target the biological mechanisms responsible for metastatic recurrence, which remains the leading cause of cancer-related mortality. The open-label, sequential dose-escalation and dose-confirmation Phase 1/2 study will evaluate the safety, tolerability, and preliminary efficacy of the investigational combination therapy in patients with relapsed or refractory sarcoma and osteosarcoma. Osteosarcoma is the most common primary bone cancer affecting children, adolescents, and young adults, and despite decades of clinical advances, survival outcomes for patients with metastatic disease have remained largely unchanged. Individuals who develop metastatic osteosarcoma continue to face five-year survival rates below 30%, underscoring the urgent need for innovative treatment strategies capable of preventing disease recurrence following initial therapy.
AI-Driven Combination Therapy Offers Novel Approach
HLX-4310 combines two complementary therapeutic mechanisms intended to interfere with the biological pathways that enable microscopic metastatic tumor cells to survive treatment and later re-emerge as life-threatening disease. The investigational regimen incorporates a class I selective HDAC inhibitor, designed to regulate gene expression and influence tumor-immune interactions, together with an mTOR inhibitor, which targets cellular pathways controlling growth, metabolism, and stress responses. Healx believes this combination may provide a differentiated strategy capable of disrupting metastatic progression before clinically detectable tumors develop. The program also represents the company’s first clinical application of its proprietary AI-enabled drug discovery platform, which integrates computational prediction, laboratory validation, and translational biology to identify promising therapeutic combinations for rare cancers and diseases with significant unmet medical need.
Expanding Innovation in Rare Oncology
The first patient dosing represents an important milestone in Healx’s expanding rare oncology pipeline and supports its long-term vision of applying artificial intelligence to accelerate precision drug development. Through collaboration with the National Pediatric Cancer Foundation’s Sunshine Projectâ„¢, the study brings together leading pediatric cancer centers to evaluate therapies designed specifically for high-risk cancers such as osteosarcoma. Researchers believe that successfully preventing metastatic recurrence in osteosarcoma could provide important scientific insights applicable to other cancers, including breast, kidney, and melanoma, where similar metastatic mechanisms are involved. As clinical development advances, HLX-4310 has the potential to establish a new therapeutic paradigm focused not only on treating cancer but also on preventing metastatic relapse, one of the greatest challenges in modern oncology and a major cause of cancer-related mortality worldwide.
