HDT Bio Corp. has launched a major Canadian clinical development initiative for HDT-401, its investigational RNA-based immunotherapy for glioblastoma, through a strategic collaboration with McGill University and The Neuro (Montreal Neurological Institute-Hospital). The program marks the establishment of HDT Bio Canada Inc., created to accelerate Canadian clinical development, regulatory activities, and biomanufacturing for the company’s oncology pipeline. Following encouraging outcomes from compassionate-use treatment, HDT-401 is advancing toward a Phase 1 clinical trial after successfully completing a pre-Clinical Trial Application meeting with Health Canada. Designed to overcome the immunosuppressive tumor microenvironment, the investigational therapy combines Riboxxim, a precisely engineered immune activator, with HDT Bio’s proprietary LIONâ„¢ nucleic acid delivery platform, representing an innovative RNA-based immunotherapy approach for one of the world’s most aggressive and difficult-to-treat brain cancers.
HDT-401 Targets Glioblastoma with Novel RNA Immunotherapy
Glioblastoma remains the most aggressive primary brain cancer in adults, with a median survival of approximately 15 months despite surgery, radiation therapy, and chemotherapy. Unlike conventional therapies, HDT-401 is designed for local administration directly into the tumor or surgical cavity, where it activates innate immune pathways and transforms the tumor environment from immune-suppressed to immune-engaged. The investigational therapy combines Riboxxim, a highly engineered TLR3/RIG-I agonist licensed from Riboxx GmbH, with the LIONâ„¢ lipid-based nucleic acid delivery platform, enabling targeted activation of inflammatory cytokines and recruitment of immune cells directly at the tumor site while minimizing systemic exposure. Encouraging observations from compassionate-use patients have helped shape the design of the upcoming Phase 1 clinical study, providing the scientific foundation for formal clinical evaluation in patients with recurrent glioblastoma.
Canadian Collaboration Accelerates Clinical Development
The upcoming Phase 1 trial will be conducted in collaboration with McGill University and The Neuro, one of Canada’s leading centers for neuroscience and neuro-oncology research. The study will be led by Dr. Roberto Diaz, together with multidisciplinary clinical and scientific experts specializing in brain tumor treatment. According to HDT Bio, the company has already completed an important regulatory milestone by holding a pre-Clinical Trial Application meeting with Health Canada, receiving constructive feedback that supports progression toward a formal Clinical Trial Application submission.
