Halozyme Therapeutics has entered into a global collaboration and license agreement with Incyte to support the development of subcutaneous formulations of INCA033989, a first-in-class mutant calreticulin (mutCALR)-targeted monoclonal antibody being investigated for myeloproliferative neoplasms (MPNs). The partnership will leverage Halozyme’s proprietary ENHANZE® drug delivery technology to enhance the subcutaneous administration of INCA033989, with the goal of improving treatment convenience, reducing patient burden, and enabling more flexible dosing regimens. In addition to the lead program, the agreement gives Incyte the option to nominate up to two additional therapeutic targets, expanding the potential application of ENHANZE® across future oncology programs. The collaboration reflects the increasing industry focus on advanced drug delivery technologies that improve patient experience while supporting the development of innovative biologic therapies.
ENHANZE® Technology Aims to Improve Subcutaneous Cancer Treatment
The collaboration centers on evaluating how ENHANZE®, Halozyme’s commercially validated drug delivery platform, can strengthen the development of INCA033989 for patients with mutCALR-expressing myeloproliferative neoplasms. ENHANZE® utilizes the proprietary rHuPH20 enzyme to facilitate rapid subcutaneous delivery of injectable medicines by temporarily increasing tissue permeability, allowing larger biologic therapies to be administered beneath the skin instead of through lengthy intravenous infusions. By incorporating this technology into Incyte’s investigational antibody program, the companies aim to develop a treatment that offers greater convenience, shorter administration times, and improved patient comfort, while maintaining clinical effectiveness. The agreement also reinforces the growing adoption of subcutaneous biologic delivery as healthcare providers increasingly seek treatment options that can reduce hospital visits and improve overall patient adherence.
Collaboration Expands Oncology Pipeline and Long-Term Partnership Opportunities
Under the financial terms of the agreement, Halozyme will receive an upfront payment from Incyte and will become eligible for additional development, regulatory, and commercial milestone payments, along with royalties on future global net sales of products utilizing ENHANZE®. Beyond the development of INCA033989, Incyte retains the option to nominate two additional therapeutic targets, creating opportunities to expand the collaboration into other oncology or hematology programs.
