SiranBio announced a major global licensing agreement with GSK for its investigational siRNA therapeutic candidate SA030, a long-acting oligonucleotide therapy targeting activin receptor-like kinase 7 (ALK7) for the treatment of metabolic and vascular diseases. Under the agreement, GSK will receive worldwide exclusive rights outside mainland China, Hong Kong, Macau, and Taiwan, with the total deal value potentially reaching US$1.005 billion through upfront payments, milestone achievements, and tiered royalties. The collaboration significantly expands GSK’s growing oligonucleotide therapeutics pipeline and highlights increasing pharmaceutical investment in next-generation RNA-based cardiometabolic therapies.
SA030 recently entered Phase I clinical trials and is being developed as a potential first-in-disease siRNA therapy designed to address cardiometabolic risk across chronic inflammatory diseases affecting the lung, liver, and kidney. The therapy specifically targets abdominal visceral adipose tissue (VAT), a major contributor to metabolic dysfunction, vascular inflammation, insulin resistance, and cardiovascular complications.
SA030 Targets High-Risk Cardiometabolic Disease
According to SiranBio and GSK, cardiometabolic disease remains the leading cause of death in nearly half of patients suffering from chronic liver and kidney disorders. Researchers believe reducing excess visceral fat while preserving lean muscle mass could significantly improve patient outcomes and survival beyond currently available therapies.
SA030 uses a differentiated fat-cell directed siRNA delivery platform developed by SiranBio to selectively target ALK7, an established biological mechanism linked to adipose tissue metabolism and inflammatory pathways. Preclinical data demonstrated the therapy’s ability to reduce abdominal fat accumulation, improve insulin sensitivity, optimize blood lipid levels, and decrease inflammation associated with chronic metabolic disorders.
The companies stated that SA030 may complement existing therapies such as GLP-1 agonists and SGLT2 inhibitors, potentially allowing future combination treatment strategies for patients with persistent cardiometabolic risk despite standard therapies. Industry experts increasingly view multi-targeted metabolic treatment approaches as critical for addressing obesity-linked cardiovascular disease, chronic inflammation, and organ fibrosis.
The program also represents an important step forward for extrahepatic siRNA delivery technologies, an area considered one of the next major frontiers in RNA therapeutics development. Unlike many traditional siRNA therapies that primarily target liver tissue, SA030 has been engineered for adipocyte-directed delivery, expanding the therapeutic potential of oligonucleotide medicines into broader metabolic disease indications.
GSK Expands RNA Therapeutics Pipeline
The agreement strengthens GSK’s strategic focus on oligonucleotide-based therapeutics, including small interfering RNA (siRNA) and antisense oligonucleotide (ASO) technologies. RNA therapeutics are increasingly being used to target disease pathways that are difficult or impossible to address using traditional small molecules or biologic drugs.
GSK executives emphasized that cardiometabolic complications remain one of the largest unmet needs in chronic inflammatory diseases involving the liver, kidney, and respiratory system. The company believes novel therapies targeting inflammation, vascular dysfunction, and fibrosis simultaneously could improve long-term outcomes for millions of patients worldwide.
Under the terms of the agreement, SiranBio will continue leading Phase I clinical development of SA030, after which GSK will assume responsibility for global development, regulatory filings, and commercialization outside the retained Asian territories. The partnership also validates SiranBio’s proprietary RNA delivery technologies, which have rapidly attracted interest from multinational pharmaceutical companies.
SiranBio stated that its broader pipeline includes innovative siRNA therapies targeting antiviral, cardiovascular, and metabolic diseases, with multiple ongoing global licensing and collaboration programs. The company’s proprietary extrahepatic delivery platforms are viewed as highly differentiated within the rapidly expanding RNA medicine sector.

The licensing agreement reflects rising pharmaceutical industry interest in RNA therapeutics for obesity, metabolic syndrome, and cardiovascular disease, some of the fastest-growing healthcare markets globally.
