SAN DIEGO, July 13, 2026
Erasca announced updated preliminary Phase 1 data for its investigational pan-RAS molecular glue ERAS-0015, demonstrating encouraging clinical activity, favorable tolerability, and continued progress toward registration-enabling development in multiple KRAS-mutant solid tumors. The company also outlined plans to initiate a potentially registration-enabling trial in second-line or later non-small cell lung cancer (NSCLC) during the first half of 2027, a Phase 3 pancreatic cancer trial in 2027, and an additional Phase 3 NSCLC study between the second half of 2027 and the first half of 2028.
ERAS-0015 Demonstrates Encouraging Responses in KRAS-Mutant Pancreatic Cancer
Updated data from the ongoing AURORAS-1 Phase 1 trial showed promising efficacy in patients with second-line or later KRAS G12X pancreatic ductal adenocarcinoma (PDAC). At the recommended dose for expansion (RDE) of 32 mg once daily, ERAS-0015 achieved an unconfirmed overall response rate (uORR) of 57% among evaluable patients. Across dose levels, patients with confirmed or unconfirmed responses remained on treatment, while most participants receiving the 24 mg and 32 mg doses continued therapy at the data cutoff, supporting the durability of clinical benefit.
Favorable Safety Profile Supports Continued Clinical Development
The updated safety analysis remained consistent with previously reported findings, demonstrating that ERAS-0015 was generally well tolerated. Most treatment-related adverse events were low grade, with no dose-limiting toxicities (DLTs), no discontinuations related to treatment, and only limited dose interruptions or reductions. Median relative dose intensity remained 100% at both the 24 mg and 32 mg dose levels, indicating that patients were able to maintain planned dosing throughout treatment.
Combination Development Progresses Toward Broader RAS-Targeted Strategy
Erasca also reported encouraging early combination data evaluating ERAS-0015 with the EGFR inhibitor panitumumab in metastatic colorectal cancer. No dose-limiting toxicities were observed during evaluation of the first 16 mg combination cohort, allowing continued dose escalation and expansion of the study. Building on these results, the company plans to further evaluate ERAS-0015 both as monotherapy and in combination regimens, with additional clinical data expected during the first half of 2027. Supported by encouraging efficacy, safety, and combination potential, Erasca is positioning ERAS-0015 as a potential foundational therapy across multiple RAS-mutant solid tumors, including pancreatic cancer, lung cancer, and colorectal cancer.
