Enodia Therapeutics has announced a research collaboration and licensing agreement with Pfizer Inc. to discover and develop novel small-molecule inhibitors targeting the Sec61 translocon, a cellular protein channel involved in transporting newly synthesized proteins. The collaboration aims to accelerate the discovery of potential medicines using Enodia’s proprietary platform for selective Sec61 modulation and targeted protein degradation. Under the agreement, the total potential value of the transaction exceeds $500 million, including an upfront payment, research and development milestones, sales milestones and tiered royalties on future net sales. The partnership brings together Enodia’s expertise in Sec61-targeting drug discovery and Pfizer’s capabilities in pharmaceutical development and commercialization. The agreement represents a significant development in research aimed at selectively targeting disease-driving proteins before they can contribute to pathological processes.
Sec61 Targeting Opens a New Drug Discovery Approach
The Sec61 translocon is a protein channel located in the membrane of the endoplasmic reticulum, a cellular structure responsible for processing and transporting many proteins. Secreted proteins and many membrane proteins rely on this pathway to reach their appropriate cellular destinations. By selectively modulating Sec61, researchers aim to interfere with the production and processing of specific disease-associated proteins at an early stage. Enodia Therapeutics is developing a proprietary platform designed to identify small molecules capable of selectively inhibiting Sec61-dependent protein transport. The approach seeks to reduce the production of harmful proteins while preserving essential physiological functions. This selectivity is a major scientific challenge because Sec61 plays a fundamental role in normal cellular biology, and broad inhibition could disrupt important processes. The company’s discovery platform combines a broad collection of small-molecule chemical structures with biological datasets, including proteomics, cryo-electron microscopy (cryo-EM) and specialized cell-line libraries. These resources support the investigation of how different molecules interact with Sec61 and how selectivity can be optimized. Machine-learning-assisted drug design is also used to guide the identification and refinement of potential inhibitor candidates. Through the collaboration, Enodia and Pfizer aim to apply this scientific approach to multiple targets, potentially expanding opportunities for the development of medicines against diseases driven by specific proteins.
Enodia and Pfizer Divide Discovery and Development Responsibilities
Under the agreement, Enodia Therapeutics will conduct the initial discovery activities, including screening, optimization and development of Sec61 inhibitor candidates against multiple collaboration targets. These activities are intended to identify and refine promising molecules for further advancement. Following the collaboration term, Pfizer will assume responsibility for clinical development, manufacturing, regulatory activities and commercialization. This division of responsibilities allows Enodia to apply its specialized discovery platform while Pfizer contributes its broader pharmaceutical development and commercial infrastructure. Enodia will receive an upfront payment and is eligible for additional research, development and sales milestone payments. The company may also receive tiered royalties on net sales if products arising from the collaboration are successfully commercialized. The agreement’s total potential value exceeds $500 million, although this amount is contingent on achieving specified milestones and future commercial outcomes. It should not be interpreted as an upfront payment or guaranteed revenue. The partnership builds on an existing relationship between the organizations. Pfizer Ventures participated in Enodia’s €20.7 million seed financing announced in January 2026. In March 2026, Enodia also acquired preclinical assets from Kezar Life Sciences related to Sec61-based drug discovery, strengthening its knowledge and capabilities in this emerging research area.
Potential Applications in Inflammation, Immunology and Oncology
Enodia’s platform is designed to support the development of small-molecule medicines for conditions involving disease-driving proteins. Its broader research focus includes inflammation, immunology and oncology, with additional potential opportunities in virology. The Pfizer collaboration will investigate multiple targets, but the companies have not publicly identified all targets or specific therapeutic indications covered by the agreement. Targeting proteins at the point of synthesis represents a different strategy from conventional approaches that inhibit a protein after it has been produced or block its activity once it reaches its site of action. If selective inhibition can be achieved, this approach could offer new opportunities to address biological targets that have been difficult to influence with existing drug-discovery methods. However, Sec61-targeting compounds remain an area of active drug research, and potential therapeutic benefits must be established through further preclinical evaluation and, where appropriate, clinical trials. Selectivity, safety and the preservation of normal protein production will be critical considerations in advancing candidates. The Enodia–Pfizer agreement highlights growing pharmaceutical interest in innovative approaches to targeted protein degradation and small-molecule discovery. The collaboration’s success will depend on identifying suitable candidates and demonstrating that they can progress through development. For cGxP.wire, the deal is a notable Bio-Pharma research and licensing milestone with potential implications for future drug development.
