ROCKVILLE, Maryland, June 2, 2026
Rise Therapeutics announced the completion of full enrollment in the four-week dose expansion cohort of its Phase 1b clinical trial evaluating R-2487 in patients with active rheumatoid arthritis (RA). The multicenter U.S. study enrolled 40 patients and marks a significant milestone in the development of the company’s oral immunotherapy candidate. Completion of enrollment strengthens the clinical dataset evaluating safety, pharmacodynamic activity, durability of response, and overall therapeutic potential, while positioning R-2487 for advancement into Phase 2 clinical development.
Expansion Cohort Supports Continued Clinical Progress
The Phase 1b trial is evaluating orally administered R-2487 in combination with standard-of-care therapies for patients with active rheumatoid arthritis. The program is specifically designed to target patients before they require treatment with biologics or JAK inhibitors, addressing an important gap in the treatment landscape. According to the company, the expanded cohort provides a broader assessment of clinical activity and reinforces the product’s positioning as a potential earlier-line treatment option. The successful enrollment milestone follows encouraging observations related to safety and durability of clinical responses, supporting continued development of the therapy in autoimmune disease.
Data Monitoring Committee Endorses Program Advancement
An independent Data Monitoring Committee (DMC) recently completed a scheduled review of available safety and clinical data from the ongoing study and recommended progression of the R-2487 program. The recommendation enables further evaluation of the therapy through extended dosing cohorts and supports preparations for Phase 2 development. Rise Therapeutics stated that data from the four-week dose expansion cohort will be presented at upcoming national and international rheumatology and immunology conferences as analyses become available. The company is also conducting additional expansion cohorts that include patients with overlapping rheumatoid arthritis and Sjögren’s disease, broadening the potential application of the investigational therapy.
Novel Immunotherapy Targets Underlying Autoimmune Dysfunction
R-2487 is a synthetic biology-based immunotherapy designed to address the root causes of autoimmune disease by inducing immune tolerance through the modulation of regulatory T cells (Tregs). Unlike traditional approaches focused primarily on immune suppression, the therapy aims to restore immune balance and potentially deliver disease-modifying benefits. The candidate is supported by Rise Therapeutics’ proprietary oral biologics delivery platform, which may offer advantages in patient convenience, treatment adherence, and earlier use compared with injectable biologic therapies. As part of its Phase 2 preparation strategy, the company is advancing dose-selection plans, regulatory engagement activities, and GMP manufacturing capabilities to support future clinical expansion. With rheumatoid arthritis representing a multi-billion-dollar global market and many patients continuing to experience limitations with existing therapies, Rise Therapeutics believes R-2487 could emerge as a differentiated treatment option for autoimmune disease management.
