Novo Nordisk has announced breakthrough Phase 3 clinical trial results for etavopivat, a first-in-class pyruvate kinase-R (PKR) activator, demonstrating significant reductions in vaso-occlusive crises (VOCs) and improved haemoglobin response in patients with sickle cell disease (SCD). Phase 3 HIBISCUS Trial Shows Strong Clinical Benefit

In the randomised, double-blind Phase 3 HIBISCUS study, involving 385 patients aged 12 years and older, etavopivat met both co-primary endpoints, demonstrating superior efficacy compared to placebo. The therapy achieved a 27% reduction in annualised VOC events, a critical outcome given that these painful crises are a defining and debilitating feature of sickle cell disease.

Additionally, time to first VOC was significantly delayed, with patients receiving etavopivat experiencing a median onset of 38.4 weeks compared to 20.9 weeks for placebo, highlighting its disease-modifying potential.
