Alpha Cognition Inc. announced positive top-line results from the BEACON Phase IV real-world study evaluating ZUNVEYL® (benzgalantamine) in long-term care patients with mild-to-moderate Alzheimer’s disease. The retrospective observational study included 162 patients treated across long-term care facilities and demonstrated favorable provider-reported outcomes in cognition, neuropsychiatric symptoms, daily functioning, medication burden, sleep quality, and gastrointestinal tolerability. While the study was not designed to establish causality, the findings provide important real-world insights into the treatment experience of Alzheimer’s patients receiving ZUNVEYL.
BEACON Study Shows Positive Cognitive and Functional Outcomes
The BEACON study collected provider assessments from 21 investigators who treated patients with ZUNVEYL for three to twelve months following prior acetylcholinesterase inhibitor therapy. Providers reported cognitive improvement in 92% of patients and favorable changes in activities of daily living (ADLs) in 71% of patients after treatment initiation. These observations suggest potential benefits in maintaining cognitive and functional performance among elderly patients living in long-term care settings, a population often underrepresented in traditional clinical studies.
Improvements Reported in Neuropsychiatric Symptoms and Medication Use
Investigators also reported improvements in neuropsychiatric symptoms in 93% of patients, including observations related to behavioral and psychological challenges commonly associated with Alzheimer’s disease. Additionally, providers indicated that 80% of patients experienced delayed initiation or discontinuation of concomitant psychotropic medications, particularly antipsychotics and anxiolytics. Although the observational design does not establish a direct causal relationship, these findings may help inform future research exploring treatment strategies aimed at reducing medication burden in complex Alzheimer’s patients.
Favorable Tolerability and Treatment Experience Observed
The study reported strong tolerability outcomes, with 98% of providers describing an overall favorable treatment experience among patients receiving ZUNVEYL. Improvements in sleep-related outcomes were reported in 72% of affected patients, while 89% of patients with prior gastrointestinal side effects from other acetylcholinesterase inhibitors were reported to have favorable gastrointestinal tolerability after transitioning to ZUNVEYL. Alpha Cognition noted that the findings reflect real-world provider observations and should not be interpreted as proof that ZUNVEYL directly caused the reported outcomes. The company believes the results contribute valuable information regarding the treatment experience of Alzheimer’s patients in long-term care environments.
