Circular Genomics has unveiled CircPATH™, its proprietary circular RNA (circRNA) discovery and validation platform, following the publication of a landmark Nature Medicine study demonstrating breakthrough performance in the early detection and prediction of Alzheimer’s disease (AD). The newly launched platform is designed to transform precision neurology by enabling blood-based detection of Alzheimer’s years before clinical symptoms appear, while significantly outperforming current gold-standard biomarkers. CircPATH™ Delivers Superior Accuracy Over Existing Alzheimer’s Biomarkers
The landmark Nature Medicine publication established that Circular Genomics’ CircPATH™ platform significantly outperformed the current clinical standard biomarker plasma pTau217 in detecting Alzheimer’s disease. The proprietary 34-circular RNA biomarker signature achieved an impressive Area Under the Curve (AUC) of 0.945 for identifying biomarker-confirmed Alzheimer’s disease in symptomatic patients, compared with 0.877 for pTau217. When both biomarker approaches were combined, diagnostic performance increased further to an exceptional AUC of 0.967, approaching the accuracy of expensive PET imaging and invasive cerebrospinal fluid testing. Beyond diagnosis, CircPATH™ demonstrated remarkable predictive capability by identifying cognitively healthy individuals who later progressed to symptomatic Alzheimer’s disease.
The circRNA biomarkers achieved a hazard ratio of 2.92, representing a 61% stronger predictive performance than pTau217 alone. For predicting symptom development within five years, CircPATH™ delivered an AUC of 0.870, substantially outperforming pTau217, which achieved an AUC of 0.676. These findings position blood-based circular RNA biomarkers as one of the most promising non-invasive technologies for identifying patients at risk before irreversible neurological damage occurs.
Multi-Pathway circRNA Signatures Advance Precision Neurology
Unlike conventional biomarkers that primarily focus on amyloid and tau pathology, CircPATH™ captures multiple biological pathways simultaneously, offering a more comprehensive picture of Alzheimer’s disease progression. The identified circRNA signatures originate from brain-enriched molecules capable of crossing the blood-brain barrier, enabling a simple blood sample to reflect complex central nervous system biology. The biomarker panel incorporates signals linked to amyloid precursor protein (APP) regulation, synaptic dysfunction, neuroinflammation, protein aggregation, and several genome-wide association study (GWAS) risk genes, including PICALM and DNAJC6. This multi-pathway approach enables more accurate disease characterization while supporting precision medicine strategies that extend beyond diagnosis.
