Caliway Biopharmaceuticals announced new preclinical data demonstrating the potential of its proprietary fat-reduction candidate CBL-514 to enhance the effectiveness of GLP-1 receptor-based weight-loss therapies and reduce weight regain after treatment discontinuation. The findings were presented at the American Diabetes Association (ADA) 2026 Scientific Sessions by obesity and metabolic medicine expert Dr. Timothy Garvey, highlighting CBL-514’s potential role in both weight management and metabolic health improvement. The data generated strong interest from international researchers and pharmaceutical companies seeking innovative approaches to obesity treatment.
Combination Therapy Significantly Reduces Post-Treatment Weight Regain
The newly presented OI25 animal study evaluated CBL-514 in combination with tirzepatide, one of the leading GLP-1-based weight-loss therapies. Results showed that the combination therapy achieved greater weight reduction than tirzepatide alone while significantly improving weight maintenance after treatment cessation. Animals receiving tirzepatide monotherapy experienced 46.1% weight regain following discontinuation, whereas those treated with the CBL-514 combination regimen recorded only 17.1% weight regain. These findings indicate that weight regain was 2.7 times higher in the monotherapy group, suggesting that CBL-514 may help extend the durability of weight-loss benefits and address one of the most significant limitations associated with current obesity therapies.
Enhanced Metabolic Benefits Beyond Weight Reduction
In addition to improved weight maintenance, the study demonstrated notable metabolic benefits from combining CBL-514 with tirzepatide. Researchers observed an additional 106.7% reduction in hepatic lipid content compared with tirzepatide alone, indicating a potential role in reducing liver fat accumulation. The combination therapy also produced meaningful improvements in the HOMA-IR index, a key marker of insulin resistance and metabolic health. These results suggest that CBL-514 may provide broader therapeutic value by supporting systemic metabolic improvements alongside localized fat reduction and weight management.
Growing Evidence Supports CBL-514’s Obesity Treatment Potential
According to Caliway, previous preclinical studies, including OI21 and OI23, consistently demonstrated that CBL-514 enhances outcomes when combined with major GLP-1 therapies such as semaglutide and tirzepatide. Across studies, the investigational therapy has shown the ability to increase weight loss, reduce subcutaneous and visceral fat, attenuate weight regain after treatment discontinuation, and improve metabolic indicators. The newly released OI25 findings further strengthen the scientific rationale for combining CBL-514 with GLP-1-based therapies and support its potential positioning within the rapidly expanding obesity treatment market.
Human Clinical Development Planned for 2026
CBL-514 is a patented first-in-class small-molecule injectable therapy designed to selectively induce adipocyte apoptosis, enabling targeted reduction of subcutaneous fat without damaging surrounding tissues. Caliway reported that more than 544 participants have been enrolled across clinical studies involving CBL-514, with completed trials meeting all primary and key secondary efficacy endpoints while demonstrating favorable safety and tolerability profiles. Based on the growing body of preclinical evidence, the company plans to initiate human clinical trials evaluating CBL-514 in combination with GLP-1 therapies during the second half of 2026, while continuing to expand its global development and licensing strategy.
