Byondis announced that it will present progress across its proprietary antibody-drug conjugate (ADC) technology platforms at World ADC San Diego, highlighting the development of its lead candidate BYON14156 and its ByonGuard™ steric masking technology. The company is using more than 20 years of in-house ADC expertise to develop novel payloads, masking approaches and linker and conjugation strategies intended to address limitations associated with current and next-generation ADC therapies. Byondis said the platform portfolio is designed to support improved efficacy, duration of response and safety in difficult-to-treat cancers while also providing opportunities for partnerships, licensing and co-development.
BYON14156 Advances Toward IND-Enabling Completion
Byondis is advancing the IND-enabling package for BYON14156 toward completion, representing a key step ahead of planned clinical trial initiation in 2027. The proprietary ADC incorporates what the company describes as a first-in-class antifolate payload and is directed against an antigen expressed across multiple tumor types. Byondis is developing the candidate specifically to address payload resistance, a challenge that can limit the activity of current and emerging ADCs. Dose-range-finding toxicity studies in non-human primates have shown a favorable therapeutic window and benign toxicity profile, according to the company, while earlier in vitro and in vivo studies demonstrated activity across multiple tumor models, including models resistant to topoisomerase and tubulin inhibitors.
ByonGuard Technology Targets Off-Tumor Toxicity
The company will also showcase ByonGuard™, its steric masking technology designed to conditionally activate antibody-based therapies and potentially improve their therapeutic index. Byondis said the technology is intended as a broad approach for reducing off-tumor toxicity associated with target-mediated activity. The platform can be applied to ADCs, T-cell engagers and other antibody-based therapeutic formats. At World ADC, Byondis Chief Scientific Officer Wim Dokter will present research examining conditionally active ADCs, including safety and efficacy findings supporting the technology’s potential to address limitations associated with conventional antibody therapies.
Byondis Expands Modular ADC Platform Strategy
BYON14156 and ByonGuard form part of Byondis’ broader strategy to advance proprietary ADC platform technologies while maintaining flexibility for independent development and strategic partnerships. The company is also developing antifolate, immunostimulatory phosphonate and dual-payload platforms alongside its steric masking technology. Byondis’ integrated discovery, development and manufacturing capabilities allow it to progress proprietary programs internally, while its modular technology approach supports potential licensing, co-development and collaboration opportunities.
