Boehringer Ingelheim has unveiled significant new oncology data at the 2026 American Society of Clinical Oncology (ASCO) Annual Meeting, highlighting promising clinical activity across multiple cancer types for its rapidly expanding precision oncology portfolio led by HERNEXEOS® (zongertinib) and investigational T-cell engager obrixtamig.
The latest results reinforce the company’s growing presence in targeted cancer therapies and immuno-oncology as it continues advancing next-generation treatments for patients with difficult-to-treat cancers including HER2-mutant non-small cell lung cancer (NSCLC), colorectal cancer, breast cancer, esophageal cancer, and extensive-stage small cell lung cancer (ES-SCLC). Zongertinib Shows Promising Results Across HER2-Driven Cancers
One of the most closely watched updates at ASCO 2026 involved new patient-reported outcomes data from the Phase Ib Beamion LUNG-1 trial, evaluating zongertinib as a first-line oral therapy for adults with HER2-mutant advanced NSCLC. According to Boehringer Ingelheim, patients receiving zongertinib experienced rapid improvements in physical functioning and symptom burden within one week of treatment initiation, with benefits sustained over time. The data also showed a favorable safety and tolerability profile with low side-effect burden and manageable adverse events, further strengthening confidence in zongertinib’s expanding clinical role within HER2-driven cancers.
The company also presented encouraging early-stage data evaluating zongertinib across additional HER2-positive solid tumors, including metastatic colorectal cancer, metastatic breast cancer, and gastroesophageal adenocarcinoma. A pooled analysis from multiple clinical trials demonstrated a confirmed objective response rate of 42% in HER2-positive metastatic colorectal cancer patients receiving zongertinib monotherapy, while combination therapy studies in gastric and breast cancers showed promising response rates and manageable safety findings. Researchers noted that no new safety signals were identified across the combination studies, supporting continued clinical expansion of zongertinib across multiple tumor types.

Boehringer Ingelheim also presented updated findings from the ongoing Phase I DAREON®-8 trial evaluating obrixtamig, an investigational DLL3/CD3-targeting T-cell engager, in combination with chemotherapy and atezolizumab for first-line treatment of extensive-stage small cell lung cancer. The updated efficacy results demonstrated a confirmed objective response rate of 73%, including complete and partial responses, while the disease control rate reached 91% among enrolled patients. Median progression-free survival and duration of response had not yet been reached at the time of data cutoff, further highlighting the therapy’s potential in aggressive neuroendocrine malignancies.
The company noted that the safety profile remained generally consistent with known treatment effects, with cytokine release syndrome identified as the most common obrixtamig-related adverse event. Researchers continue to view DLL3-directed therapies as one of the most promising emerging approaches for treating small cell lung cancer, an area with limited long-term therapeutic advancements.
