Antengene Corporation Limited has announced a landmark strategic licensing agreement with K2 Therapeutics, a biotechnology company established by MPM BioImpact, granting exclusive global rights outside Greater China for the development and commercialization of ATG-106, a first-in-class CDH6 x CD3 bispecific T-cell engager (TCE) currently in preclinical development for solid tumors. The transaction further strengthens Antengene’s position as a leading innovator in next-generation immuno-oncology and validates the commercial potential of its proprietary AnTenGager® platform. Under the agreement, Antengene will receive approximately USD 20 million in upfront and near-term consideration, while remaining eligible for up to USD 960.5 million in development, regulatory, and commercial milestone payments, in addition to tiered royalties on future global sales. The companies also entered into an option agreement that could provide K2 Therapeutics exclusive rights to a second undisclosed preclinical bispecific TCE candidate, creating the potential for another milestone package worth up to USD 960.5 million.
Strategic Partnership Expands Global Reach of AnTenGager® Platform
The agreement represents a major commercial validation of Antengene’s proprietary AnTenGager® TCE 2.0 platform, which was specifically designed to overcome the safety and efficacy limitations that have historically restricted the success of first-generation T-cell engagers in solid tumors. The platform incorporates innovative steric hindrance masking technology and proprietary CD3-binding sequences with rapid on/off kinetics to activate T cells in a disease-targeted manner while minimizing the risk of cytokine release syndrome and T-cell exhaustion. ATG-106 leverages this technology by targeting CDH6, a protein overexpressed in multiple cancers including ovarian and renal tumors while exhibiting limited expression in healthy adult tissues.
This differentiated approach is expected to improve therapeutic precision and broaden the applicability of TCE therapies across a range of difficult-to-treat malignancies. The transaction also marks the second major commercial agreement involving an AnTenGager®-derived asset following Antengene’s global licensing agreement with UCB for ATG-201 earlier in 2026, highlighting increasing industry confidence in the platform’s potential.
